The genomes of cyanobacteria contain homologous genes of mitochondria or bacterial complex I, which encode the subunits of NAD(P)H dehydrogenase (NDH) complex participating in the cyclic electron transport, respiration and inorganic carbon concentration mechanism. However, so far, since the entire active NDH has not yet been isolated and purified, the biochemical properties,function and structure of NDH are still remained to be elucidated. The aim of this project is to investigate the biochemical properties, functions and the structure of NDH-1L isolated from a thermophilic cyanobacterium (Thermosynechococcus elongatus BP-1) in our lab recently.The project will provide theorical base for the elucidate the function of NDH in photosynthesis by demonstrating the electron imput entry, the substrate binding characters and tridimentional structure.
蓝藻基因组中存在编码与线粒体或细菌复合体I同源的基因(NDH),其基因产物所形成的复合体称为NAD(P)H脱氢酶(NDH)复合体,主要参与循环电子传递、呼吸作用、无机碳的浓缩机制。然而,由于完整的活性NDH的分离纯化尚未成功,NDH的生化性质与结构还有待于阐明。本项目以本研究组最近分离纯化得到的嗜热聚球蓝藻NDH的一种复合体NDH-1L为研究材料,拟进行NDH-1L的生化性质、功能和结构的研究,以揭示NDH-1L的电子输入结构域、底物结合特征、以及三维结构,为阐明NDH在光合作用中的作用提供理论依据。
