The mRNA in the cell body of neurons can be transported to their dendrites and axons, and translated locally to regulate neuronal functions. MicroRNA (miRNA), an important regulatory factor, has been shown to localize in the dendrites and axons of neurons and involve in important physiological functions. However, the distribution, function, mechanism and transport mode of miRNAs need to be further investigated in different neurons. The techniques including immunohisto-/cytochemistry, fluorescence in-situ hybridization, molecular cloning, dual luciferase report system, cell transfection, Western blotting, micro-injection of cell and live cell imaging etc., will be performed in the present study. We will clarify the distribution and function of axon-enriched miRNAs in primary sensory neurons, detect their down-stream target molecules, identify the long-distance transport mode of miRNAs in axons, and find the factors and signaling pathways to regulate the expression level and axon transport of miRNAs. The present study will provide important evidences for the function and mechanism of axon-enriched miRNAs.
神经元胞体中的mRNA可以运输至其树突和轴突,通过局部翻译来调控神经元的功能。作为转录后翻译的重要调节因子,MicroRNA(miRNA)已经被发现存在于神经元树突和轴突中,参与重要的生理功能,但对于不同miRNA在不同类别神经元树突和轴突中的分布、功能、作用机制和转运模式有待于深入研究。我们成功建立了初级感觉神经元微流小室分隔培养系统,筛选了相对富集于轴突内的miRNA。本课题将应用免疫组织/细胞化学、荧光原位杂交、分子克隆、双荧光素报告系统、细胞转染、免疫蛋白杂交、细胞显微注射以及活细胞影像等技术,阐明富集于初级感觉神经元轴突内miRNA的分布和功能,检测其下游作用的靶分子,揭示miRNA在轴突中的长距离转运模式,发现调控miRNA表达和轴突运输的外界因子和信号通路。本研究将为miRNA轴突定位的功能和机制提供重要的证据。
该项目主要研究初级感觉神经元轴突富集的miRNA及其功能和作用机制。建立了进行轴突中RNA和蛋白检测的研究装置和实验方法,筛选了初级感觉神经元轴突富集的miRNA,阐明了轴突富集的miR-181d及其下游靶基因对初级感觉神经元轴突生长的影响,揭示了RNA结合蛋白FMRP介导miR-181d及其靶基因向轴突的转运,发现了神经生长因子NGF信号通路对FMRP和miR-181d介导的靶基因局部翻译和轴突生长的调节作用。同时,系统地总结了神经元轴突中定位的miRNA以及在轴突发育中的功能和调节机制。研究成果在《Cell Reports》、《Journal of Molecular Cell Biology》和《Molecular Pain》杂志上发表3篇论文,在国际学术会议上做专题报告2次,在国内外学术会议上做墙报展示4次。毕业博士研究生1名,出站博士后1名,培养副研究员1名。
