The Osteoporosis often occurres in senile patients. The main Traditional Chinese Medicine syndrome type of osteoporosis is kidney and spleen deficiency and blood stasis.Lycium Barbarum L has kidney essence, nourishing liver and improving eyesight Pharmaceutical effect.Recently, we and other groups found that the major ingredient Lycium Barbarum Polysaccharides(LBP )had therapeutic effectiveness in OP. But its study only focused on morphology and biochemical index and very few study on the mechanism of the signaling pathway. Wnt signaling pathway plays an important role in regulating bone mass、bone mineral density and bone remodeling and the new target for treating OP was found. Therefore, in this study we will use both in vivo and in vitro methods including two-dimensional gel electrophoresis, Western Blot、mRNA chip、RT-PCR, electronmicroscope scan and two energy X-ray absorption to investigate the effect of LBP on OP and the protein and gene expression in Wnt signaling pathway.This study will provide experimental basis for the use of LBP in treating OP.Effects and mechanism of LBP on Wnt signaling pathway in senile osteoporosis
老年性骨质疏松症(OP)是人体机能自然衰退、老化的重要表现。中医认为OP为肾虚、脾虚和血癖。枸杞具有补肾益精、养肝明目等药效。近年来,国内外学者及本项目组研究均显示其主要成分枸杞多糖(LBP)在治疗OP方面有疗效,但多集中在形态学及生化指标改变上,对OP信号通路研究较少。在OP相关信号通路中,Wnt通路在调节骨量和骨重建中发挥重要作用,且在Wnt通路上发现治疗OP新靶点。为此本项目选用LBP为防治OP药物、以Wnt通路为切入点,利用双向凝胶电泳、基因芯片、RT-PCR、Westernblot、双能X线吸收等方法进行体内、外实验①LBP干预不同月龄SAMP6小鼠,观察骨密度等形态学指标;②不同浓度含LBP血清干预体外培养的成骨和骨髓间充质干细胞;③检测体内、外成骨和骨髓间充质干细胞Wnt通路中蛋白和基因表达变化,旨在探讨LBP对老年性OP的作用机理。
