Xinjiang is a high incidence area of bone hydatid disease, bone hydatid disease which is characterized with“mouse cave”and like cancer of infiltrating growth in bone matrix, it grows furtively, high disability and mortality rate. At home and abroad the current treatment method of bone hydatid is surgical excision, which is easily recurrent or can not eradicate, drug therapy is ineffective to bone hydatid disease, these are the mainly clinical difficult problem. Though in our pre-study, we found that radiotherapy showed to be effective on bone hydatid disease,but the security still needs to be verified, the mechanism of outcome is still unknown. This study aimed to make a systematic study about how hydatid cyst of bone is necrotic and absorbed, ulcerated and eliminated from the body, recurrent and infected. After radiation therapy in different radiation dose, explore and describe the molecular networks weather chromosomes of hydatid cells be broken, the heterogeneity of echinococcus and bone cells, the necrosis and reconstitution of bone; and further to analyze bone hydatid’s prognosis and molecular mechanism in different radiation doses, clear prognosis and evaluate the security and effectiveness of radiation therapy of bone hydatid disease, build and improve the security of the radiotherapy system, and provide guidance for the clinical application of radiation therapy of bone hydatid disease.
新疆是骨包虫病高发区,骨包虫在骨质中形同“老鼠洞”样的蔓延酷似癌性浸润性生长,生长隐秘,致残和致死率很高,临床上易误诊误治、易复发难以根治。目前国内外对骨包虫治疗以手术切除为主,易复发,无法根除,药物治疗无效,这些是其主要的临床难题。我们前期研究显示放射治疗骨包虫有一定疗效,但其安全性仍需验证,其转归如何仍未知。本研究拟通过对前期放射治疗后骨包虫动物模型病变部位骨包虫囊坏死、吸收、破溃排出体外及复发、感染等开展系统研究,描绘不同放射剂量下照射后出现包虫细胞染色体断裂和包虫囊/骨细胞接受放射线的的异质性及骨骼坏死/重建的分子网络,解析接受不同放射剂量后骨包虫的转归及其分子机制,明确预后评价放射治疗骨包虫病的安全性和有效性,构建和完善安全的放疗体系,为放射治疗骨包虫病的临床应用提供指导。
新疆是骨包虫病高发区,骨包虫在骨质中形同“老鼠洞”样的蔓延酷似癌性浸润性生长,生长隐秘,致残和致死率很高,临床上易误诊误治、易复发难以根治。目前国内外对骨包虫治疗以手术切除为主,易复发,无法根除,药物治疗无效,这些是其主要的临床难题。本研究对放射治疗后骨包虫动物模型病变部位组织病理改变以及病变组织Ca2+和蛋白质含量改变的系统研究。研究过程中对骨包虫建模方法进行了改良。结果表明50Gy剂量的放射治疗是对沙鼠骨包虫病有抑制和治疗作用的安全剂量。电离辐射干扰Ca2+稳态的机制复杂,是IR治疗骨包虫病的机制之一。本研究通过解析接受不同放射剂量后骨包虫的转归及其分子机制,明确预后评价放射治疗骨包虫病的安全性和有效性,构建和完善安全的放疗体系,为放射治疗骨包虫病的临床应用具有参考意义。
