In the current field of reproduction,it has been one of the hotspots and difficult issue to explore fertility preservation strategies for women with tumors. For patients with various types of tumors, it will be the best way to extract preantral follicles from cryo-preservation ovarian tissue in vitro development and maturation. All the current research at home and abroad has showed that the process of maturation in vitro for human preantral follicles is hindered and the oocytes are kept low quality as influenced by environment, support, and many other factors. However, there is no a good culture system to solve the problem at present. Recently, our research team has innovatively constructed a new and better DNA hydrogel 3D culture system without enzymatic digestion and better cell proliferation when compared with the alginate, hyaluronic acid or collagen protein 3D culture system. At the meantime, our team also found that HUMSCs can promote the process of maturation of immature oocyte in vitro and improve the oocytes quality. We will comprehensively validate DNA as the best hydrogel by comparing the four kinds of frequented-used 3D culture system, and explore the mechanism of the new 3D culture system with HUMSCs/DNA hydrogel in the role of preantral follicle development in vitro, and then to build a new type of 3D culture system for human preantral follicles in vitro development and provide a new strategy for the research on female fertility preservation.

探索女性肿瘤患者生育力保存策略是目前生殖领域研究的热点和难点之一。肿瘤患者卵巢组织低温保存并获取窦前卵泡体外培养至成熟将是解决肿瘤患者生育力问题的最佳手段。目前国内外研究均显示,窦前卵泡体外发育过程中,受微环境、支撑力等诸多因素的影响,卵母细胞发育成熟受阻且卵母细胞质量低下,然而目前缺乏一种较好的培养体系来解决这些问题。本课题组创新性地构建了DNA水凝胶3D培养体系,并初步发现其相对优于藻酸盐、透明质酸和胶原蛋白3D培养体系;同时,本课题组发现HUMSCs可以增进未成熟卵母细胞的体外成熟和质量。本项目在前期研究基础上深入,拟通过对比四种水凝胶3D培养体系在窦前卵泡体外发育中的作用,全面验证DNA水凝胶为构建3D培养体系的最佳水凝胶,并探索HUMSCs联合DNA水凝胶所形成的3D培养体系的作用及机制,从而构建一种新型的有利于窦前卵泡体外发育的3D培养体系,为女性生育力保存研究提供新策略。
