KDR/flk-1基因在胃肠道间质瘤中的突变及突变型基因功能的研究

本项目以胃肠道间质瘤（gastrointestinal stromal tumor，GIST）临床病理及c-kit基因突变的研究为基础，以研究中发现的新线索KDR/flk-1基因可能与GIST有关为切入点，拟采用免疫组织化学、RACE-PCR以及基因序列测定等方法研究KDR/flk-1基因蛋白产物VEGFR2在GIST中的表达状况，确定KDR/flk-1基因在GIST中的突变位点及突变规律，进一步鉴定突变型KDR/flk-1基因生物学功能。探讨KDR/flk-1基因在GIST发生发展中的作用以及蛋白产物VEGFR2在GIST中表达的意义，从分子水平达到对GIST更深入的认识，同时为GIST的辅助治疗提供研究基础。
