CNTF gene modified neural stem cell transplantation to play an important role in the repair of optic nerve injury, but its the rapeutic mechanism is still a lack of in-depth study. Our previous study found that the CNTF gene modified neural stem cells of the protective effect of optic nerve injury; observe to the miRNA-134, LIMK-1 after optic nerve injury expression differences; pursuant to which the proposed the CNTF gene-modified neural stem cell transplantation by sustained expression of ciliary neurotrophic factor, inhibit the expression of the miRNA-134, the increase of the expression of LIMK-1 to promote the repair of optic nerve injury. We intend to use the nerve bundles Road track, neurophysiological, immunohistochemical and gene chips, protein two-dimensional electrophoresis and other molecular biology to observe the migration, differentiation of transplanted cells in vivo and post-transplant gene expression differences to explore the CNTF gene-modified neural stem cell transplantation promote optic nerve injury repair mechanisms. This study will elucidate the molecular mechanisms of the CNTF gene modified neural stem cell transplantation to promote repair after optic nerve injury, has important significance, and will provide new ideas for the treatment of optic nerve injury.
CNTF基因修饰神经干细胞移植在视神经损伤的修复中起重要作用,但其治疗机制尚缺乏深入研究。我们前期研究发现CNTF基因修饰神经干细胞对损伤视神经的保护效应;也观察到视神经损伤后miRNA-134、LIMK-1表达的差异;据此提出CNTF基因修饰神经干细胞移植通过持续表达睫状神经营养因子,抑制miRNA-134的表达,增加LIMK-1的表达而促使受损视神经的修复。我们拟采用神经束路追踪、神经电生理、免疫组化及基因芯片、蛋白质双向电泳等分子生物学方法观察移植细胞在体内的迁移、分化和移植后基因表达差异,探讨CNTF基因修饰神经干细胞移植促进视神经损伤的修复机制。本研究对阐明CNTF基因修饰神经干细胞移植促进视神经损伤后修复的分子机制有重要意义,并将为视神经损伤的治疗提供新的思路。
