Our previous study confirmed for the first time that LncRNA HOTAIR was highly expressed in glioma cells, and and closely related with the prognosis of glioma patients. In further study, we found that HOTAIR 3' and 5' functional domains play a different role in regulation of glioma cell cycle progression. Results showed that HOTAIR regulate glioma cell cycle progression mainly through its 5' domain in an EZH2 dependent manner...The current experimental results of our group suggest that HOTAIR also play an important role in glioma cell invasion, but the specific molecular mechanism is unknown. The aim of this study is to explore the functions of different domains of HOTAIR in the regulation of glioma cell invasion through establishing 3' domain/5' domain over-expressed glioma cells. We will use the techniques of coimmunoprecipitation, RNA immunoprecipitation and chromatin immunoprecipitation in our study. ..In this study, the molecular mechanism of HOTAIR regulation of glioma invasion will be described in detail, which provides a new molecular target for the treatment of glioma.
我们课题组前期实验结果首次证实长链非编码RNA HOTAIR在胶质瘤细胞中呈高表达,并且与胶质瘤患者的预后密切相关。在进一步的研究中,我们发现HOTAIR的3’及5’功能结构域在其调控胶质瘤细胞周期进展中发挥不同的作用,HOTAIR主要通过其5’功能结构域以EZH2依赖性的方式调节胶质瘤细胞周期进展。.目前本课题组正在进行的实验结果提示HOTAIR在胶质瘤侵袭中也发挥了重要作用,但具体分子机制不详。本课题将在前期实验的基础上,通过构建过表达HOTAIR 3’及5’功能结构域的胶质瘤细胞为模型,利用蛋白质免疫共沉淀、RNA免疫沉淀、染色质免疫沉淀等技术探索HOTAIR的不同功能结构域在调控胶质瘤侵袭中发挥的作用。.本研究可详细阐述HOTAIR调控胶质瘤侵袭的分子机制,为临床治疗胶质瘤提供新的分子靶标。
