Lung cancer is one of the malignant tumors with the highest incidence and mortality rate. Ferroptosis has recently been discovered as a type of cell death, which is expected to become a new target for cancer treatment. There are few studies on ferroptosis and lncRNA. Therefore, it is important to find out the regulation mechanism between ferroptosis and lncRNA. Previous studies found that NR_033957 was low-expressed in lung adenocarcinoma and associated with poor prognosis, inhibiting the progression of tumor cells by promoting ferroptosis. Based on these findings, we will conduct the following studies: (1) To explore the abnormal low expression level of NR_033957 in lung adenocarcinoma and its regulation mechanism; (2) Confirm the biological function of NR_033957 in vivo and in vitro; (3) To screen and identify the downstream target genes and interaction proteins of NR_033957. This study will elucidate the molecular mechanism of NR_033957 promotes lung adenocarcinoma ferroptosis, and provide new targets and scientific basis for the treatment of lung adenocarcinoma.
肺癌是发病率与死亡率最高的恶性肿瘤之一。铁死亡是近年发现的一种细胞死亡方式,有望成为肿瘤治疗新靶标。目前关于铁死亡与lncRNA的研究较少,因此寻找铁死亡相关lncRNA并探讨其调控机制具有重要意义。申请人前期研究发现,NR_033957在肺腺癌中表达水平异常降低并与患者预后不良相关,体内外实验发现其可通过促进铁死亡发生来抑制肿瘤演进。基于此,我们拟开展以下研究内容:(1)探讨NR_033957在肺腺癌中表达异常及其机制;(2)体内外实验证实其生物学功能;(3)筛选NR_033957调控的下游靶基因和相互作用蛋白并验证。本项目拟阐明NR_033957促进肺腺癌细胞发生铁死亡的分子机制,为肺腺癌治疗提供新的靶点和依据。
