Immune regulation between mater and fetus during pregnancy, especially periimplantation, is one of core objects of reproductive immunology, which is very important to comprehensive the nature of pregnant regulation in animals and improve the rate of embryo transfer and animal reproduction. On the basis of studying the activation of CD58 on EML and PBMC in goats , this programm was further performed with the methods MTT and ELISA, as followed: 1. The effect of CD58 on the cytokines such as IL-2, IL-4, TGF-βs, secreted by EML and γδT cells; 2. The kinetics of the above cytokines and IFN-τin uterine liquid during early pregnancy ; 3. The effect of CD58 in vivo on the above cytokines in uterine liquid during early pregnancy; 4. the relationship between CD58 and EPF. The results are that CD58 can regulate the immune balance in the local uterine, tranform the balance among cytokine net into the good state for fetus development, and improve the para-immune by effecting the activation of EML and γδT cells and secretion of the above cytokines. Another, a character of antigen cross between CD58 and EPF is proved. On the basis of all above results, the immunologic regulation of CD58 in ruminant during early pregnancy is primarily elucidated, which develops the model' CD58 regulation of the local-uterine cytokine during early pregnancy', and builds a milestone for understanding the immunologic mechanism in the local-uterine cytokine during early pregnancy, and using CD58 to enhance pregnant rate and carry out pregnant diagnosis. In addition to, the effect of Selenium(Se) on activating the above cells and secreting cytokines is performed. The following conclusions are drawn: Selenium can accelerate the activation of goat EML and PBMC in vitro in a dose-dependent manner; Selenium through oral administration can decrease the level of IL-2 significantly, and increase significantly the level of IL-4,TGF-β1 and TGF-β2 during early pregnancy in goat. In short, it demonstrates firstly that selenium can regulate the reproductive immunity, especially the local-uterine cytokine network during early pregnancy in goat. It enriches the theories that selenium can be used to prevent early embryo death and improve the reproductive function of animals.
分别应用MTT法、ELISA及免疫印迹等方法测定分析山羊妊娠第9d-10d子宫液中CD58、ILs、TGF-B及IFN-t的动态规律,CD58对分泌IFN-t及其mRNA表达的影响CD58对妊娠早期子宫局部细胞因子网络的调节作用及其机制,对全面认识子宫局部免疫调节及其与胚胎发育的关系有重要理论意义。为其在提高胚胎移植成功率和防治早期胚胎死亡等方面的应用提供理论依据。
