Live cancer is one of the most common malignant tumors in the world, and it has threatened people’s health seriously. The precision diagnosis and prognosis evaluation of liver cancer is the only way to improve curative effect and prognosis, and it is also a major clinical problem to be solved urgently. This project is established in the unique value of multiphoton microscopy in the “pathological and biological characteristics” diagnosis and prognosis evaluation of diseases. In this project, a new multiphoton microscopy of liver will be developed by the coupling of intrinsic two-photon excited fluorescence (TPEF), and second-harmonic generation (SHG) modalities. The intrinsic signals of liver for this new technique will be investigated, and the effects of new system parameters on multiphoton imaging depth and contrast will be studied to acquire the optimal system parameters. This new technique will be applied to study on the precision diagnosis and prognosis evaluation of liver cancer. This project will try to explore the quantitative link between intrinsic multiphoton signals and liver cancer biological characteristics, invasion, metastatic potential, prognosis. In summary, this project will offer a new tool, some useful evidences and experimental data for the precision diagnosis and prognosis evaluation of liver cancer quantitatively. This project also will provide novel ideas and theoretical support for individual therapy of liver cancer. It is of great academic significance and clinical value.
肝癌是世界范围内最常见的恶性肿瘤之一,严重威胁人类的健康。实现肝癌的精准诊断和预后预测是提高疗效和改善预后的唯一途径,也是目前迫切需要解决的重大临床问题。本项目瞄准多光子显微技术在疾病“病理-生物学”诊断及预后评估的独特价值,发展基于内源性光信号的肝组织多光子显微新技术,通过对肝癌精准诊断和预后评估等科学问题的研究,揭示新技术的成像机理与制定新技术的最佳系统参数,建立光学诊断信息与肝癌生物学特性之间的定量关系,探究内源性多光子信号与肝癌转移潜能、预后的相关性。预期成果将为肝癌精准诊断和预后评估提供直观、可量化的参考依据和技术手段,为实现肝癌精准治疗,提高肝癌患者治愈率与生存率提供新思路和理论支持,具有重要的学术意义和临床价值。
