建立了TRAP-ELISA法检测了15株消化系肿瘤细胞,13株端粒酶阳性(包括肝癌细胞株SMMC-7721、BEL-7402),表明绝大多数肿瘤细胞表达端粒酶活性。用此方法检测良恶性腹水端粒酶活性,发现肝硬化、结核性腹膜炎腹水脱落细胞不能检测端粒酶活性,而癌性腹水(肝癌、胃癌、结肠癌、胰腺癌、卵巢癌)85.29%端粒酶阳性,特异性和敏感性优于腹水AFI、CEA、CA系列,敏感性高于腹水细胞学检查,端粒酶活性检测在良恶性腹水鉴别诊断中具有较高的临床价值。设计合成针对端粒酶RNA成分的反义DNA,观察到该反义DNA可以显著抑制肝癌细胞端粒酶活性并抑制肝癌细胞增殖,诱导细胞凋亡,此作用与三氧化二砷抗肝癌作用的协同反应。
