The blood stasis has been supposed to be the important pathogenesis of erectile function (ED), and the changes of vascualr endothelial cells (VECs) prove to be its crucial element. Increasing evidence suggest that activating blood circulation dissipates blood stasis and protecting or rebuilding endothelial cellular function through different pathways. However, due to lack of ideal experimental ED animal models with blood stasis syndrome and three dimensional cell culture system which integrate disease and syndrome in vitro, the mechanism of activating blood circulation to regulate corpus cavernosum endothelial cell function remains not to be completely clarified. The project is designed to first build a kind of remarkable and meaningful ED animal model with blood stasis syndrome and a three dimensional endothelial cell culture system which integrate disease and syndrome, based on a significant effect of activating blood circulation on endothelial cellular function restore in ED patients with blood stasis syndrome, and then apply high throughput signal trasduction function microarray and downstream verification techniques, to explore the possible mechanism of activating blood circulation to restore corpus cavernosum endothelial cell function for more experimental and theoretical evidences for traditional Chinese therapy in ED treatment.
血瘀是勃起功能障碍(ED)的重要病机,血管内皮细胞功能变化是血瘀证病变的重要环节,活血化瘀法能狗保护和修复内皮细胞功能,但由于缺乏理想的血瘀证ED动物实验模型,其对血瘀证及活血化瘀法对阴茎海绵体内皮细胞结构及功能的具体影响至今尚未阐明。本项目拟通过建立具有创新意义的血瘀证ED动物模型,探究血瘀证及活血化瘀法对阴茎海绵体内皮细胞结构及功能的具体影响,为ED中医中药治疗提供实验、理论依据。
