Rho associated coiled coil forming protein kinase 2(Rock2) is closely associated with tumorigenesis and metastasis. Our study found that Rock2 could play protective effect on liver cancer cell survival exposing to ionizing radiation (IR) induced DNA damage and Rock2 also could regulate cell cycle checkpoint. Simultaneously,we also found that Rock2 protein phosphorylation occurs and the Rock2 serine 1121 site occurred phosphorylation using specific phosphorylated antibodies exposing to IR induced DNA damage.But The upstream kinase of Rock2 phosphorylated point and the regulatory role in DNA damage repair pathway are still unclear exposing to IR induced DNA damage. This object is intended to be used by methods such as, gene-directed mutagenesis, laser scanning confocal and flow cytometry to determine Rock2 phosphorylation upstream receptors, to observe the distribution of the association of Rock2 and Rock2 ser-1121 site and DNA damage sites in liver cancer cells, to study the biological activity on liver cancer cells.We will identify the impact of Rock2 on DNA damage repair pathway, further clarify the relationship with primary carcinoma of liver and provide theoretical basis for the development of primary carcinoma of liver.
Rho相关卷曲螺旋激酶2(Rock2)与肿瘤的发生、转移等有密切关联。我们的研究发现电离辐射(IR)引起DNA损伤时Rock2对肝癌细胞的生存具有保护作用,而且Rock2对肝癌细胞周期检查点有调控作用。同时,我们还证实IR诱导肝癌细胞DNA损伤时,Rock2在丝氨酸1121位点发生磷酸化,并用特异性磷酸化抗体加以验证。但IR诱导DNA损伤时Rock2磷酸化位点在DNA损伤修复通路中的调控作用及Rock2磷酸化的上游作用激酶尚不清楚。本课题拟运用基因定点突变、激光共聚焦及流式细胞术等方法确定Rock2磷酸化的上游感受器,观察肝癌细胞中Rock2及磷酸化位点与DNA损伤位点分布情况的关联,并分析IR诱导DNA损伤时Rock2的修复方式及对肝癌细胞生物学活性的影响,以明确Rock2在DNA损伤修复通路中的作用,进一步阐明Rock2与肝癌的关系,并为肝癌的发生发展机制研究提供理论基础。
肝癌发病机制复杂,广泛的信号通路对肝癌的发生发展起着重要作用。目前国内外对肝癌的发生机制研究仍是热点。对于 Rock2 的研究,国外研究发现其在许多不同的肿瘤组织如肝癌、食道癌、膀胱癌、睾丸癌、胰腺癌等中均观察到 Rock2 基因表达的增高,并且还与其他许多恶性表型相关。有研究提出 Rho /Rock 通路可以做为肿瘤治疗的靶点。而且,研究表明肝癌细胞中降低 Rock2 表达可以降低其侵袭转移能力;反之,增加 Rock2 的表达,可以增强肝癌的侵袭转移能力。.本课题研究阐明了Rock2在电离辐射诱导DNA损伤修复通路中的重要作用,确定了电离辐射诱导DNA损伤时Rock2磷酸化上游作用激酶ATM,了解了Rock2磷酸化位点Ser1121与DNA损伤位点γ-H2AX的关联,分析出Rock2磷酸化位点Ser1121对DNA同源和非同源修复的调控作用,进而探明了Rock2磷酸化位点Ser1121对肝癌细胞生物学活性的影响。本课题将为进一步了解Rock2在肝癌放射敏感性中的作用提供了理论依据,为肝癌的发生发展机制研究提供新的研究靶点,最终为肝癌的临床治疗提供新的思路。通过本课题的研究,将会促进在原发性肝癌发生发展机制中的研究,提高科研水平,推动肝癌探索的临床及教学发展,为肝癌的诊治提供坚实的理论基础。
