The placenta plays a vital role in many aspects of fetal development during pregnancy. Dysfunction of placenta caused by oxidative stress can lead to the developmental abnormality of the fetuses. So, academic and application value will be brought in with the development of effective antioxidants to deal with the oxidative stress of the placenta. In some researches, tryptophan and its metabolites have been reported to have efficient ability to clean free radicals. So we speculated that tryptophan and some tryptophan metabolites may act as potent antioxidants in the placenta. But so far, researches about the antioxidant properties of tryptophan and the mechanism of preventing oxidative stress of it in the placenta are still very few. In this project, modern molecular biological, biochemical, cell biological techniques and omics technology are integrated to systematically study the antioxygenation of tryptophan and HA in pig placental trophoblastic cells. And then we try to explore the metabolic and molecular mechanism of the antioxygenation of tryptophan in placental trophoblastic cells. At the same time, these studies can apply theoretical basis and technical support for the using of tryptophan to regulate the oxidation resistance of the pregnant sow and enrich the theory of nutritional values of amino acids in pig.
胎盘发生氧化应激,严重时引起胎盘功能障碍,影响胎儿的正常发育。开发有效的抗氧化剂来缓解胎盘内的氧化应激具有重要的学术及应用价值。色氨酸及其代谢产物具有高效的抗氧化性。本项目综合采用细胞生物学和分子生物学等技术,以猪胎盘滋养层原代培养细胞为研究对象,系统地研究色氨酸及代谢产物3-羟基氨基苯甲酸在胎盘滋养层细胞中的抗氧化功能。同时,以mTOR信号通路和Nrf2/ARE信号通路为切入点,探究色氨酸影响滋养层细胞抗氧化性的分子机制,为胎盘抗氧化剂的开发提供有效的验证模式和靶标基因。本研究旨在为色氨酸添加调控妊娠母猪胎盘抗氧化性提供理论基础和技术支撑,同时丰富猪的氨基酸营养理论。
色氨酸是哺乳动物体内必需氨基酸和重要的功能性物质,具有多种生物学功能。色氨酸及其代谢产物具有高效的抗氧化性,但色氨酸在胎盘组织中抗氧化作用并未做研究,同时,还缺乏色氨酸、色氨酸代谢在影响胎盘抗氧化性的整体认识。本项目成功建立胎盘滋养层细胞氧化应激模型;获得色氨酸、褪黑素、5-羟色胺、犬尿氨酸和3-羟基2氨基苯甲酸影响猪胎盘滋养层细胞抗氧化性的有效作用浓度;初步揭示了色氨酸影响胎盘滋养层细胞增殖及犬尿氨酸、褪黑素及3-羟基2氨基苯甲酸影响胎盘滋养层细胞抗氧化性的分子机制。已有数据发表SCI论文8篇。该项目结果为,为色氨酸作为妊娠母猪抗氧化性饲料添加剂提供理论支撑。
