Screening arsenism-related genes by using mRNA differential display PCR technique. After comparision the different gene expression between normal human skin tissues and arsenism patient skin tissues, we acquired arsenism disease-related gene fragments sequence and their selective expression in normal human skin tissues and arsenism patiens/skin cancer patients. Sequence analysis showed that some of these disease related gene fragments are known genes while some are unknown genes. The results were further confirmed by Northern blot analysis. The analysis of the gene expression changes has important significance in discovery of the mechanism of arsenism and arsenism related cancer.
用mRNA差异显示技术筛选地方性砷中毒相关基因,获得新新基因全序列及染色体定位,或已知基因在正常组织及病变、癌变组织中的选择性表达,探讨砷中毒在分子水平的发病和至癌机理,特别是癌基因抑癌基因的表达水平变化,对于砷中毒机理的阐明和对地方性砷中毒防治问题的根本解决具有重要的意义。.
