Long noncoding RNA MALAT1 is closely associated with the invasion and metastasis of esophageal cancer. However, the mechanism of MALAT1’s effect on invasion and metastasis of esophagus carcinoma remains to be elucidated until now. MALAT1 may form a complex with c-Jun based on our preliminary results. Besides, there are a bunch of overlap genes between MALAT1 and c-Jun. Thus, this program will validates the interaction sites between MALAT1 and c-Jun, interrogates MALAT1’s effects on c-Jun relocation in esophagus carcinoma genome, and unravel the regulation mechanism of MALAT1 -c-Jun, lncRNA-transcription factor complex, on regulation of invasion and metastasis related genes in esophagus carcinoma. The outcome of this program is expected to give a better understanding of esophagus carcinoma invasion and metastasis, which is of great scientific value.
长链非编码RNA(long noncoding RNA,lncRNA)MALAT1与食管癌侵袭转移密切相关,但目前其调控食管癌侵袭转移的分子机制仍不清楚。本项目前期研究发现,在食管癌中MALAT1能够与c-Jun相互作用;且MALAT1与c-Jun存在多个共同调控的下游基因。基于此,本项目拟进一步确定MALAT1与c-Jun相互作用位点,并探究MALAT1对c-Jun在食管癌基因组定位的影响,最终揭示MALAT1-c-Jun lncRNA-转录因子复合物调控食管癌侵袭转移的分子机制。相关结果对于食管癌侵袭转移机理的深入理解具有重要的科学意义。
