bFGF is a important growth factor during the process of wound healing. The role of bFGF in scar formation is still unclear. It's also a disputed question of bFGF receptor's expression in hypertrophic scar and normal skin as well as its distribution in fibroblasts. In this study, Immunohistochemistry, flow cytometer and western-blot were used to investigate the differential expression of FGFR1 and its subcellular expression in hypertrophic scar and normal skin and in normal and hypertrophic scar's fibroblasts. In our study we found that FGFR1 positive cells were observed in keratinocytes, sweat and sebaceous gland cells, vescular endothelial cells and fibroblasts. the expression of FGFR1 in fibroblasts was similar between hypertrophic scar and normal skin and is mainly located at nuclear and lessly in cytoplasmic. This study indicated that bFGF plays an important role in promoting wound healing in original period of wound healing process. However, in the process of hypretrophic scar's formation, bFGF may not be a key factor.
增生性疤痕是现代医学的难题之一。本课题对照研究正常皮肤和增生性疤痕成纤维细胞中成纤维细胞生长因子受体(FGFR)的亚细胞分布,并用转基因技术提高FGFR的表达,观察FGFR对成纤维细胞增殖及胶原合成的影响,探讨预防和治疗增生性疤痕的可能途径。此项研究国内外未见报道。研究成果对肝纤维化、矽肺等纤维化疾病也具有重要意义。.
