Oligoasthenospermia has long been the most discussed subject and remains as a problem in andrology. Huang Jing Zan Yu Capsule Capsule, a kind of spermatogenic prescription developed by our team for the treatment of sterility , has demonstrated positive therapeutic effect clinically. Based on the rationale of “ Spectrum-effect relationship” of readily-made Chinese medicine and new findings on the role of SCF/c-kit signal pathway in regulating the generation and apoptosis of sperm cells, it is hypothesized that active components in Huang Jing Zan Yu Capsule is likely to be a cure for Oligoasthenospermia throughout its regulation on the SCF/c-kit signal pathway. Aided with information technology,HPLC-DAD and HPLC-ELSD fingerprinting are established to investigate the regulatory effect of Huang Jing Zan Yu Capsule to mice with oligoasthenospermia. The “ Spectrum-effect relationship” of Huang Jing Zan Yu Capsule is also developed based on our previous research findings, with an attempt to pinpoint the valid target for treatment for oligoasthenospermia and its action mechanism,providing reference for clinical rational use of drug and guiding the treatment of infertility with the use of direct traditional Chinese medicine.
少、弱精子症是男性不育领域研究的热点和难题。课题组前期以黄精赞育胶囊干预少、弱精子症取得了良好的临床疗效。本研究根据中药复方“谱-效”关系理念,结合SCF/c-kit信号通路调控生精细胞凋亡机制的最新认识,提出假说:黄精赞育胶囊通过有效成分谱调控SCF/c-kit信号通路的整体功能治疗少、弱精子症。拟采用信息融合技术建立黄精赞育胶囊体外指纹图谱与体内血清药物化学相结合的HPLC-DAD和HPLC-ELSD指纹图谱;并检测黄精赞育胶囊对少、弱精子症大鼠SCF/c-kit信号通路的整体变化。整合以上研究结果,建立黄精赞育胶囊调节SCF/c-kit信号通路的“谱-效”关系,旨在科学阐释黄精赞育胶囊治疗少、弱精子症的有效靶点和作用机制,为临床合理用药提供依据,对于指导中医药治疗男性不育具有重要意义。
课题组前期以黄精赞育胶囊干预少、弱精子症取得了良好的临床疗效。本研究通过体外化学成分及体内药物血清移行组分分析,建立黄精赞育胶囊体外化学成分HPLC的测定方法、MSn的测定方法,获取黄精赞育胶囊化学成分及体内移行成分,为质控研究提供依据,为药效机制研究奠定基础。建立少弱精子症大鼠模型,探讨黄精赞育胶囊干预模型大鼠的精液质量、内分泌激素、睾丸病理形态学及超微结构、调亡相关因子Caspase-3、Bax和Bcl-2蛋白及mRNA表达的变化,以及睾丸基因表达谱的改变等。为黄精赞育胶囊的药效物质基础和分子作用机制研究提供支撑,并为黄精赞育胶囊的组方用药及作用机制提供科学内涵。研究发现五味子甲素是黄精赞育胶囊体内重要入血成分,并对睾丸间质细胞增殖具有促进作用。通过五味子甲素对少、弱精子症大鼠模型进行干预,观察其各项精液参数、组织形态以及性激素的变化,明确五味子甲素改善少、弱精子症模型的作用效果。通过分子生物学实验技术,探究五味子甲素干预后SCF/c-kit信号通路凋亡相关因子表达水平的变化。结合五味子甲素对形态学及分子层面上的改变,验证其抑制生精细胞凋亡的作用机制,为进一步揭示五味子甲素改善少、弱精子症大鼠作用原理提供思路。少、弱精子症可通过对睾丸组织形态学的改变、内分泌紊乱、促凋亡基因的过表达以及抗凋亡基因的抑制致使精子数量减少、质量下降,最终导致男性不育的发生。五味子甲素可通过恢复生精小管的形态及功能,改善睾丸内环境,上调抗凋亡基因SCF表达以抑制SCF/c-kit信号通路产生的异常凋亡,从而达到生精的目的,是黄精赞育胶囊治疗少、弱精子症的有效成分之一。SCF/c-kit信号通路在调控生精细胞凋亡的同时也调控细胞增殖分化。整合以上研究结果,建立黄精赞育胶囊调节SCF/c-kit信号通路的“谱-效”关系,为临床合理用药提供依据,对于指导中医药治疗男性不育具有重要意义。
