Innate lymphoid cells (ILCs) are a growing family of immune cells that mirror the phenotypes and functions of helper T cells. Unlike T cells and B cells, ILCs do not have antigen-specific receptors. As the main sources producing IL-22, ILC3s can promote tissue repair and maintain homeostasis. We have been reported the hepato-protective role of IL-22 during drug induced liver injury (DILI) and chronic hepatitis B. To the best of our knowledge, no research on ILCs in DILI has been reported in the field of DILI. This project proposes to investigate the frequency and function of ILCs in DILI patients with different inflammatory phrases by using peripheral blood and liver tissues from patients. Specifically, we further employ an adoptive transfer approach with ILC3 which uses Rag1 knockout and IL-22 knockout mice and explores the role of ILC3 and IL-22 in the development of DILI. The works in our group connect the ILC3 and its produced IL-22 in the development of DILI, thus further expanding the study of the immune mechanism of liver injury. The mechanism of ILC3 and IL-22 in DILI provides new immunotherapeutic strategies for treating DILI.
固有淋巴细胞(innate lymphoid cells, ILCs)是一类新近发现、具有与辅助性T细胞相似表型和功能的“镜像细胞”, 但缺乏T、B细胞的抗原特异性受体。作为IL-22的主要来源,3型ILC细胞(ILC3)具有组织修复、重塑及维持组织内环境稳定的功能。我们前期工作发现IL-22在药物性肝损伤(DILI)及慢性乙型肝炎中发挥肝保护效应,但目前尚无ILCs在DILI中的研究报道。本项目拟分析DILI患者不同炎症期ILC3细胞在外周血中的频数及肝内的浸润程度;应用过继转移ILC3的方法,选用Rag1基因敲除小鼠、IL-22基因敲除小鼠研究ILC3细胞与IL-22在DILI病程中的作用机制,将ILC3细胞及其分泌的IL-22与DILI的修复联系起来,并以此为视窗,进而拓展肝损伤的免疫学机制研究,为DILI的免疫治疗策略提供新的思路和方法。
固有淋巴细胞(ILCs)是一类具有与辅助性T细胞相似表型和功能的“镜像细胞”, 但缺乏T、B细胞的抗原特异性受体。研究发现,作为IL-22的主要来源,3型ILC细胞(ILC3)在组织修复、重塑及维持组织内环境稳定方面发挥重要功能。 我们前期工作发现IL-22在药物性肝损伤(DILI)及慢性乙型肝炎中发挥肝保护效应,但尚无ILCs在DILI中的研究报道。在本课题中,我们首先在药物性肝损伤患者的外周血中发现ILC3细胞频数明显降低,显著低于健康对照组,通过随访发现,其细胞频数与疾病转归相关,但与转氨酶、总胆红素等肝生化指标无关。分泌IL-22的外周血ILC3细胞频数改变在DILI的早期预警信号中可能存在潜在价值。在APAP诱导的急性肝损伤小鼠模型中,外源性重组IL-22蛋白通过激活AMPK通路依赖的自噬活化,进而减少氧化应激及肝内炎性细胞因子的表达,显著改善APAP诱导的小鼠肝损伤。
