The health of women in pregnancy is not only important to the mother herself, but also has a long-term impact on the health of the whole life of the next generation. At present, gestational diabetes mellitus (GDM) lacks the consideration of gut microbiota in clinical nutrition interventions, and gut microbiota plays an important role in the process of human intake, degradation, absorption and metabolism of nutrients. By sequencing the gut microflora of GDM and normal pregnant women with 16S RNA sequencing, we found that there were significant differences in the level of the phylum, the genus. This subject intends to carry out the metabolic pathway analysis among the two groups of 8 genus gut microbiota, find out the genetic information difference of gut microbiota associated with glucose metabolism of the host. By using the downstream products for intervention on pregnant GDM mice, to validate the mechanism of gene affected by the glucose metabolism of the host. The completion of this subject can provide a theoretical basis for the clinical prevention of glycometabolism disorder in pregnancy, and the improvement of the current clinical nutrition intervention measures for GDM pregnant women.
妇女妊娠期的健康不仅关系母亲自身,而且对下一代整个生命阶段的健康都具有长远影响。目前的妊娠糖尿病(GDM)孕妇的临床营养干预措施中缺少了对肠道菌群的考虑,而肠道菌群在人体摄入营养的降解,吸收和代谢过程中起到重要的作用。我们通过对GDM孕妇和正常孕妇的肠道菌群16s RNA测序分析后,发现了在门,属以及菌种水平都有明显差异。本课题拟对两组人群差异明显的8个菌种进一步做代谢通路模拟分析,得出可能与宿主糖代谢相关的肠道菌群差异基因信息,并利用差异基因表达下游产物对GDM孕鼠进行干预,以验证分析得到的基因影响宿主糖代谢的机制。本课题的完成可为临床预防妊娠期糖代谢紊乱,以及完善目前的GDM孕妇临床营养干预措施提供理论依据。
妇女妊娠期的健康不仅关系母亲自身,而且对下一代整个生命阶段的健康都具有长远影响。目前的妊娠糖尿病(GDM)孕妇的临床营养干预措施中缺少了对肠道菌群的考虑,而肠道菌群在人体摄入营养的降解,吸收和代谢过程中起到重要的作用。本项目从GDM孕妇特征性肠内菌的分析出发,得出GDM孕妇糖代谢紊乱与孕期特定肠内乳杆菌减少和特定短肽代谢产物减少相关;GDM孕妇分娩后肠内菌类黄酮合成功能基因多于正常孕妇,与其通过胞内cAMP降解甘油三酯的通路活跃相关。此结果可用于今后研发GDM临床精准营养,解决患者在孕期既需要控制血糖又需要加强营养的难题。
