A novel functional heart valve has been developed by using tissue engineering technique to transplant active cells onto biodegradable scaffolds.The seeded cells proliferate,organize,and produce cellular and extracellular matrix.During this matrix formation,the starter scaffolds is degrade,absorbed,or metabolized.This type of heart valve has the optimal abilities in biocompatibility,anticoagulation and longevity.The seeded cell growth and hypodermic implantation of collagenous film and polyer of polygycolic acid/polyactic acid(PGLA) were compared.Collagenous film retained the membranous structure 8 weeks after hypodermic implantation,and complete degradation of the polymer occurred by 8-12 weeks.The biocompatibility was better than that of PGLA.Conclusions Collagenous film can be used to fabricate a three-dimensional,biodegradable heart valve scaffold by tissue engineering.
现有的人工生物瓣与机械瓣膜各有其优缺点及其应用的局限性,同种瓣膜应用受到来源限制。因此采用组织工程学技术研制开发自体细胞心脏瓣膜不仅有重要学术价值,而且有广泛应用前景。研究内容:①筛选一种天然可吸收材料作为瓣膜基质支架,将自体细胞种植其上制成瓣膜;②组织工程瓣膜移植动物体内,观察生物相容性,基质支架吸收和自射组织取代结果。
