Miiuy croaker, Miichthys miiuy is an important commercial marine farming fish that is distributed in the East China Sea; the increasingly serious bacterial diseases hinder the development of this species aquaculture industry, however, the basic immunology research still lags behind of miiuy croaker. In our previous study of more than 20 immune-related genes revealed the molecular mechanisms involved in the immune response, and described the regulation of gene expression occurred in response to foreign pathogens, but it is not yet clear the molecular mechanism of the regulation. The project intends to make use of genomics data to carry out the study of the molecular mechanism of immune-related miRNAs. In order to obtain: 1) Joint genome data to build a comprehensive and detailed miiuy croaker miRNAs family and to elaborate molecular evolutionary mechanisms of the miRNAs clusters of immune-related miRNAs; 2)Screening and identification immune-related miRNAs and target genes. Carry miRNAs-mRNA interaction studies and reveal the mechanisms of post-transcriptional regulation; 3) Identification the transcription factor of immune-related miRNAs and elucidate transcriptional regulation mechanism, preliminary clarity on the regulation of downstream signaling network; 4) To carry out the functional verification of miRNAs and the upstream and downstream factors regulating the performance of the body's resistance to disease, and to lay the theoretical foundation for further development of molecular immune mechanisms and immune regulation of disease resistance molecular breeding research.
鮸鱼是东海海域重要的养殖经济鱼种,日益严重的细菌性疾病阻碍了养殖产业的发展,而该物种的相关基础免疫学研究还相对滞后。我们前期对20多个抗病免疫相关基因的研究初步揭示了先天性免疫及适应性免疫基因参与免疫应答的分子机制,并阐述了应对外来病原所发生的基因表达调控过程,但尚不明晰其调控的分子机制。本项目拟利用组学数据开展免疫相关miRNAs分子调控机制的研究:1)基于基因组数据构建全面详尽的鮸鱼miRNAs谱系,比较阐述鱼类免疫相关miRNAs家族簇的分子进化机制;2)筛选并鉴定免疫相关miRNAs及靶基因,开展miRNAs-mRNA互作研究并揭示转录后调控机制;3)阐明免疫相关miRNAs的转录调控因子及转录调控机制,初步明晰上下游信号调控网络;4)开展miRNAs及上下游因子调控机体抗病性能的功能验证,为进一步开展分子免疫机制及免疫调控抗病分子育种研究奠定理论基础。
鮸鱼是东海海域重要的养殖经济鱼种,日益严重的细菌性疾病阻碍了养殖产业的发展,而该物种及养殖鱼类的相关分子免疫学研究还相对滞后,亟需要深入开展分子免疫学的相关研究。本项目以鮸鱼为研究对象,开展了免疫组学及miRNA分子调控机制的系列相关研究,主要研究内容如下:1)基于鮸鱼全基因组、转录组和small RNA转录组数据,全面详尽的研究了鮸鱼miRNA表达谱,比较阐述了鱼类免疫相关miRNAs家族簇的分子进化机制;2)筛选并鉴定了一批免疫相关miRNAs及其靶基因,开展了miRNAs-mRNA互作研究并揭示了转录后调控机制;3)阐明了一批重要的免疫miRNAs的转录调控因子及转录调控机制,初步明晰上下游信号调控网络。该项目的完成为进一步开展分子免疫机制及免疫调控抗病分子育种研究奠定理论基础。
