Sweetpotato Virus Disease (SPVD) is a globally devastating disease in main sweetpotato producers. Owing to resistant germplasm and genetic study being rare, it seriously hinders sweetpotato SPVD-resistant breeding and genes molecular linked marker. Through the last NSF’s carrying out, several SPVD-resistant wild species had been introduced and identified. We also succeeded to obtain two interspecific hybrids F1 and first generation BC1F1. Based on these works, the proposal was put forward to construct interspecific BC2F1 introgression lines (ILs) resistant to SPVD by following backcrosses and SSR molecular markers. And we also study how the alien genome and chromosome fragments or genes was introgressed into or made recombination with receptor’s genome.
甘薯病毒病(Sweetpotato Virus Disease, SPVD)是危害甘薯生产的一个全球性重大病害之一。由于栽培种内抗源不足和抗病遗传研究滞后,严重影响了甘薯抗病毒育种和优异抗性基因标记和定位。通过上个国家自然科学基金的实施,引进、鉴定和筛选了多份对SPVD具有极端抗性的野生资源,成功获得种间杂种F1和BC1F1群体。本项目在此基础上进一步渐渗回交,重点筛选一批抗/较耐SPVD优异株系,再回交并通过SSR标记辅助建立BC2F1渐渗系(Introgression lines, ILS),为建立高代渐渗系和构建饱和遗传连锁图谱奠定基础。研究初级渐渗外源染色体组、片段甚至基因与受体亲本染色体组渐渗和重组变化规律。
甘薯病毒病是危害甘薯生产的一个全球性重大病害之一。通过本项目实施,构建了BC2F1渐渗群体122株,表型鉴定分析和SSR分子标记分析表明此群体遗传结构合理,抗SPVD变异分化明显,适于抗性遗传分析。从中筛选到了抗病株系9株,耐病株系19株。发表论文2篇,1篇会议论文,另已撰写1篇论文正在投稿。申请国家发明专利一项(申请号:201410121827.8.)。参加国际会议两次和国内会议一次。培养硕士生一名。
