A gene of VBP is isolated firstly from Myzus persicae.When virus particles actively move in hemocoael of aphid vector for virus transmission it plays a role of protection for virus. The VBP gene,1.6kb,encodes 548 amino acids,and possesses 92.0% homology related to seqence of groEL gene,thus,the gene to code VBP in aphid is identified as groEL.Furthermore,other three VBP genes also are isolated from Rhopalosiphum padi,Rhopalosiphum maidis, Schizaphis graminum.The sequences of VBP genes in Myzus persicae and Rhopalosiphum padi exist with 98% homology and a differentiation of 3 amino acids,which may play a important role in circulation process of aphid vectors. The VBP genes are expressed into a 63 ku protein in vector pBV221,and the 69 ku fusion protein in vectors pET30a and pTricHisA respectively. The VBP has a key function in protection of virus form degradation in hemocoael by binding virus particles during circulation process of aphid vectors with ultramicroscopy by using immuno labeled VBP antibody made in rabbit.The mechanism of virus trnansmissions in stylet-borne and circulative nanner are analysed.
作物蚜传病毒病数目很多,极易爆发流行成灾。本研究首次开展桃蚜体内病毒结合蛋白基因的克隆及全序列分析,探讨蚜虫传播专化性的分子基础,以及与形成蚜虫口针传播和循回传播方式间的关系。为利用该基因防治病毒病提供科学依据。本研究既是本学科重要的理论问题,也是我国及全球性作物生产中此类病害防治面临的急待解决的严重问题。
