Effective prevention and treatment of radiation-induced brain injury has always been an important and urgent problem in clinical radiation therapy. Our early work has identified that magnesium can protect the radiation-induced brain injury.Recent studies found the NF-κB/ ICAM-1 pathway had played an important regulating role in injury of vascular endothelial cells.Our early studies have also identified magnesium can effectively reduce the expression of ICAM-1 in brain tissue and the injury of brain vascular through regulating NF-κB/ ICAM-1 pathway.For further study upon the molecular mechanism of magnesium prevention and treatment on radiation-induced brain injury,this work will focus on the regulating mechanism of the NF-κB/ ICAM-1 pathway in the levels of cells and animals.In addition,our results will provide experimental basis for clinical application of magnesium.It will be an important guide to reduce radiation-induced brain injury and improve the control of tumor radiation therapy.
有效地防治放射性脑损伤一直是临床头颈部放射治疗亟待解决的重要问题。本课题组已有研究证实镁离子对放射性脑损伤具有保护作用。近年来研究发现,NF-κB/ICAM-1通路在血管内皮细胞损伤中发挥着重要的调控作用。本课题组前期研究结果表明,镁离子可有效地降低放射引起的脑组织中ICAM-1的表达,并减轻脑血管的损伤。这可能是通过NF-κB/ICAM-1通路发挥的调控作用。为深入探讨镁离子防治放射性脑损伤的分子机制,本课题拟从细胞和动物水平详细研究镁离子通过NF-κB/ ICAM-1通路防治放射性脑损伤的调控机制,研究结果不仅阐明NF-κB/ ICAM-1通路对放射性脑损伤的调控作用,还为镁离子最终在临床放射治疗上应用提供详实的生物学实验依据,对改善放射性脑损伤和提高肿瘤放疗的效果具有重要的指导意义和应用价值。
有效地防治放射性脑损伤一直是临床头颈部放射治疗亟待解决的重要问题。近年来研究发现,硫酸镁注射液对放射性脑损伤后的脑水肿具有很好的防治作用,此外,放射性脑损伤后早期应用硫酸镁还可以改善远期神经功能障碍。但硫酸镁具体的分子机制一直不明确。本项目主要研究内容:硫酸镁对人脐静脉血管内皮细胞(HUVEC)放射损伤的防护作用及其分子机制;硫酸镁对小鼠放射性脑损伤时血管内皮细胞防护作用的分子机制。研究结果:硫酸镁降低HUVEC照射后产生的自由基且经NF-κB/ICAM-1调节HUVEC细胞的放射损伤,降低HUVEC照射后的DNA损伤,缓解HUVEC照射后的细胞周期阻滞,促进细胞凋亡;5MeV 20Gy的电子束照射能成功建立小鼠的放射性脑损伤模型,300mg/kg浓度的硫酸镁能够明显的改善放射性脑损伤的病理状况;硫酸镁通过抑制NF-κB基因的表达,从而使得ICAM-1的表达降低,通过NF-κB/ICAM-1信号通路对小鼠放射性脑损伤起到保护作用。本项目从细胞和动物水平详细阐明镁离子通过NF-κB/ICAM-1通路防治放射性脑损伤的调控机制,为镁离子最终在临床放射治疗上应用提供详实的生物学实验依据,对改善放射性脑损伤和提高肿瘤放疗的效果具有重要的指导意义和应用价值。
