High myopia is one of the world's major irreversible blinding eye disease.In the process of development of high myopia , abnormal extension and thinning of the posterior sclera , associated with excessive degradation of scleral extracellular matrix, is the one of the main reasons for the development of high myopia.The current Western medicine has no effective treatment, and common medical and surgical therapy has uncertain effects and big toxic side effects, which are not suited to use widely. We use traditional Chinese medicine Nourishing blood and tonifying kidney Decoction to treat high myopia, and found it can improve patients's visual function. Its mechanism remains to be studied further. Jak/Stat signal transduction pathway may lead to an important pathway of scleral extracellular matrix remodeling.The lens-induced high myopia model is used to do the research. Light microscopy, electron microscopy, flow cytometry, immunohistochemistry and Western blotting are used to detect the apoptosis of scleral cells and the expression of MMP-2, TIMP-2, p-Stat3. So we can observe the Intervention effect of the TCM on high myopia, investigate the signal transduction pathway of high myopia scleral extracellular matrix remodeling and explor the mechanism in cellular and molecular levels. This study will provide experimental basis for the protective effect of traditional Chinese medicine on high myopia, and has significant sense to prevent high myopia.
高度近视是目前全球主要不可逆致盲性眼病之一,高度近视的发展过程中眼球后部巩膜异常扩展和变薄,伴有巩膜细胞外基质的过度降解是高度近视不断发展的根本病因,目前西医对此尚无有效的治疗方法,常用的药物及手术疗法疗效不确且毒副作用大,不适合推广使用。我们临床采用中药养血补肾方治疗高度近视,能改善患者的视功能,其机理尚待进一步研究。Jak/Stat信号转导通路可能是导致巩膜细胞外基质的重塑的一条重要通路。本课题应用透镜诱导高度近视动物模型,采用光镜、电镜、流式细胞术、免疫组化、蛋白印迹等方法检测巩膜细胞的凋亡、MMP-2 及TIMP-2、p-Stat3的表达变化,观察养血补肾方对该模型的干预作用,从分子生物学的角度探讨影响高度近视巩膜细胞外基质重塑的信号转导通路及养血补肾方防治高度近视的可能机理。本研究将为中药对高度近视巩膜的保护作用提供实验依据,对高度近视防治取得突破性进展具有深远意义。
