Male infertility, recurrent abortion is closely related with the sperm DNA damage. Sperm miRNA is very important in the process of spermatogenesis, early embryonic development and sperm DNA damage. Environmental pollutants can change the expression of miRNA in sperm, which can lead to infertility, abortion, etc. Based on the previous research foundation, the research group proposed that "Some Male infertility, recurrent abortion is closely related with the sperm DNA damage. Sperm miRNA is very important in the process of sperm DNA damage, spermatogenesis and early embryonic development. Environmental pollutants can change the expression of miRNA in sperm, which can lead to infertility, abortion, etc. Based on the previous research foundation, the research group proposed that "Some exogenous miRNAs in Yishen Tongluo Decoction have the protective effect on the sperm DNA damage ". To verify the hypothesis, the internationally recognized environment pollution of benzene and (a) pyrene (BAP) poisoning animal model will be established, and treated with Yishen Tongluo Decoction. The sperm DNA integrity and miRNAs expression of parental and filial generations will be detected using related experimental technology. To screen the related protective miRNAs of the sperm DNA damage. Detect the existence of the corresponding miRNAs of Yishen Tongluo Decoction, drug containing serum and testicular tissue. To solve the protective exogenous miRNAs on the sperm DNA in the Yishen Tongluo Decoction , and which miRNAs through the blood testis barrier. It will provide conclusive evidence for traditional Chinese medicine protecting the genetic material damage caused by environmental pollution.

男性不育、复发性流产与精子DNA损伤关系密切。精子miRNA在精子DNA损伤、精子发生及胚胎早期发育过程中至关重要。环境污染物可影响精子miRNA表达从而导致不育、流产等。植物中的某些外源性miRNA可经消化道吸收稳定存在于血液中,影响某些重要基因的表达。课题组基于前期研究基础,提出“益肾通络方中的某些外源性miRNA可能具有防护精子DNA损伤的作用”的假说。本课题拟通过复制国际公认环境污染物苯并(a)芘(BaP)染毒小鼠模型,并以益肾通络方进行干预,对亲代及其子代小鼠应用相关实验技术检测精子DNA完整性及miRNA表达谱,筛选得出益肾通络方防护精子DNA损伤的相关miRNA;检测益肾通络方水煎液、含药血清及睾丸组织中是否存在相应miRNA,拟解决益肾通络方中所含防护精子DNA的外源性miRNA,以及透过血睾屏障的miRNA有哪些。为中医药防护因环境污染物所致遗传物质的损伤,提供确切证据。

项目摘要

本课题通过复制国际公认环境污染物苯并(a)芘(BaP)染毒大鼠模型,并以益肾通络方进行干预,应用相关实验技术检测精子DNA完整性及miRNA表达谱,筛选得出益肾通络方防护精子DNA损伤的相关miRNA;检测益肾通络方水煎液、含药血清及睾丸组织中是否存在相应miRNA,筛选得出了益肾通络方中所含防护精子DNA的外源性miRNA。为中医药防护因环境污染物所致遗传物质的损伤,提供确切证据。
