Recurrent spontaneous abortion (RSA) is often defined as two or more consecutive miscarriages, which has complicated pathogenesis. However, few treatment exist, especially treatment that target improving biological function of trophoblast cells. In this study, we aim at investigating the effect of human umbilical cord mesenchymal stem cells (hUCMSCs) on invasion ability of trophoblasts, and on pathogenesis of RSA as well. In our previous study, we found that hUCMSCs have a positive effect on HTR-8/SVneo trophoblast cell lines’ biological functions including proliferation, migration, invasion and secretion. Since dysfunction of trophoblasts’ invasion ability plays significant role in pathogenesis of RSA, in this project we will further study the effect of hUCMSCs on invasion ability and hCG secretion ability of primary cultured trophoblasts, and of explant cultured placental tissues, and its possible mechanism. Meanwhile, we establish RSA animal model by mating combination CBA/J×DBA/2J mouse , and give hUCMSCs treatment to experiment groups, and then calculate the ratio of abortions of each groups. This study might provide a new avenue for treatment of trophoblast-related diseases like RSA.
复发性流产是指与同一性伴侣连续发生2次或2次以上自然流产,其病因及发病机制非常复杂,且目前并无稳定的治疗或预防方法,尤其是针对胎盘滋养细胞侵袭功能障碍的措施还很少,其研究意义巨大。本课题旨在研究人脐带间充质干细胞通过改善滋养细胞的侵袭能力等生物学功能,从而对复发性流产的病理生理过程产生良性的影响。我们的前期研究发现,人脐带间充质干细胞可通过共培养或上清干预两种方式促进HTR-8/SVneo滋养细胞系的增殖、迁移、侵袭和分泌功能,而滋养细胞的侵袭能力异常在复发性流产的发病机制中具有重要意义,本课题将继续深入研究人脐带间充质干细胞对原代培养滋养细胞以及体外培养的绒毛组织的侵袭功能、hCG分泌功能的影响,及其可能机制;同时利用雌性CBA/J小鼠与雄性DBA/2小鼠交配建立自然流产模型,实验组给予hUCMSCs治疗,比较胚胎吸收率的差异。以期为复发性流产等滋养细胞相关妊娠疾病的治疗提供新的思路。
