五指山猪矮小性遗传机理的研究

Using Pit-1 gene as candidate gene for dwarfism in this project. The Pit-1 gene was screened for genetic variation in three Chinese miniature pig breeds: Wuzhishan, Diannan Small-ear, Xiangzhu and three Chinese common pig breeds: Laiwu Black, Beijing Black, Yimeng Black by PCR-SSCP, Microsatellite DNA and sequence scan method. The result indicates: three PCR-SSCP genotypes were found in the Exon 4 of Pit-1 gene in Wuzhishan, Laiwu Black, Beijing Black, Yimeng Black pigs and a single nucleotide of T→C at position 316 in Wuzhishan, Xiangzhu pigs. We can find correlation between genetic variation and dwarfism. The genetic variation study was also on above three Chinese miniature pig breeds by using ten microsatellite loci.

以Pit-1基因作为矮小性能的侯选基因，利用PCR-RFLP微卫星DNA等分子标记技术，研究五指山猪和普通猪Pit-1基因内的遗传变异，分析Pit-1基因在小型猪中的遗传特性，探讨猪矮小性的遗传机理，为确立近交系WZSP分子遗传学特异性监测标准提供理论依据。
