EST-SSR as a new kind of molecular marker has been developed and used in many fruit trees because it has several advantages over genomic-derived SSRs, such as easy and inexpensive to develop, high transferability, besides possessing the features of the latter one. Although the molecular and genetic research of peach as a model of perennial woody fruit crops has been made more progress, little attention has been paid to the potential transferability of peach EST-SSRs to other Rosaceae relatives. In this study, we intend to develop and analysis of peach EST-SSRs from network databases with abundant ESTs. EST-SSRs of peach which authenticity and polymorphism were identified were used to analysis of phylogenetic relationship with germplasm resources. The development of peach EST-SSRs was to provide effective molecular markers for genetic diversity analysis, genetic linkage map construction, molecular marker assisted breeding and gene location of peach. Meanwhile, the transferability of peach EST-SSRs was investigated in 45 individual members of the Rosaceae family representing four genera. Some of the EST-SSRs proved transferable to other species in the Rosaceae providing additional markers for comparative mapping, development of molecular markers and discovery of homologous gene among the Rosaceae family.
EST-SSR标记作为一种新型的SSR标记,不仅具有传统基因组SSR标记的特点,而且以其开发过程简单、成本低廉及在物种间通用性较高等优势已在多种果树中得到开发和应用。桃作为多年生木本果树进行分子遗传研究的模式植物,其分子遗传研究进展较快,但关于桃EST-SSR引物在蔷薇科中通用性的研究却鲜有报道。本研究拟利用公共网络数据库中丰富的ESTs数据进行桃EST-SSR标记的开发和分析,并将经过真实性验证的具有多态性的EST-SSR引物应用于桃种质资源亲缘关系分析,具有多态性的桃EST-SSR标记的开发为进一步的桃遗传多样性分析、遗传图谱构建、分子标记辅助育种、重要基因定位提供有效标记;此外,根据不同物种间EST-SSR引物具有通用性,将已开发的桃EST-SSR标记应用于蔷薇科不同种属其他果树,检测其在其他果树的通用性,从而为蔷薇科其他果树分子标记开发和不同物种间比较作图及同源基因发掘提供新途径。
EST-SSR标记以其开发过程简单、成本低廉及在物种间通用性较高等优势已在多种果树中得到开发和应用。桃作为多年生木本果树进行分子遗传研究的模式植物,其分子遗传研究进展较快,但关于桃EST-SSR引物在蔷薇科中通用性的研究却鲜有报道。.本项目针对这一研究背景,主要进行以下四个方面的研究,研究结果如下:.1.从NCBI数据库中获得桃EST序列83837条,利用Phrap软件对这些ESTs进行拼接,获得30756条序列。其中含SSR的EST有2911条,包含单核苷酸重复676条,二核苷酸重复1673条,三核苷酸重复277条,四核苷酸重复86条,五核苷酸重复133条,六核苷酸重复102条。在所获得的SSR重复单元中,AG/CT和AT/AT在二核苷酸中占主导地位;三核苷酸重复基元中比例较大的为ATC/ATG;四核苷酸重复基元中AAAT/ATTT和ATCC/ATGG出现频率较高;五核苷酸重复基元中AAGGC/CCTTG出现频率最高;55种六核苷酸重复基元出现频率均低于6.00%。.2.利用Primer Premier 5.0软件设计桃EST-SSR引物528对,以6个桃种质为实验材料进行引物有效性扩增验证,其中可扩增出目的条带的引物有298对,产生多态性条带的有174对。将50对具有多态性的桃EST-SSR引物在6个桃样品中扩增位点的DNA片段分别回收进行测序分析,结果显示所有扩增产物的SSR基序与EST中的SSR基序相同,但是相同引物在不同品种间基序重复次数不同。.3.选择40对具有多态性的桃EST-SSR引物对桃6种生长型的10个桃种质和其五个变种的12个种质进行分析,筛选出12对在桃不同树型及其变种间具有较高的多态性的桃EST-SSR标记。.4.随机选取的30对具有多态性的桃EST-SSR引物和37对基因组SSR引物分别在蔷薇科4个属共72个品种中进行扩增,结果表明大部分桃EST-SSR和基因组SSR引物均能在在蔷薇科其他果树中扩增出目的条带;其中30对桃EST-SSR引物在苹果、梨、杏、李、草莓和樱桃中扩增的多态性比率分别为76.00%、72.00%、85.19%、85.19%、65.22%和93.10%;而37对桃基因组SSR引物在苹果、梨、杏、李、草莓和樱桃中扩增的多态性比率分别为76.00%、89.19%、89.19%、83.78%、91.89%和94.59%。
