Allergic rhinitis(AR),as a commonly diaease in the field of otorhinolaryngology,still have an unclear mechanism on pathogenesis . microRNA(miRNA) is a group of gene regulator in human immune response,which plays important role on the differentiation,development and regulatory of immune cells.Recently,research showed that different subsets of γδT cell participate in the Th2 type immune response,and each has opposite regulatory function.In this study,regulation of γδT cells by microRNA was designed as the entry point,we aim to study the regulation of microRNA on γδT cells and its role on the development of allergic rhinitis by the expression of the subset of γδT cells.This study may determine the microRNA expression profile of γδT cell and the balance of γδT/Treg in allergic rhinitis .The study may fill the blank of the research between microRNA and allergic rhinitis ,and the pathogenesis too.The proposed study will potentially provide a new target for treatment in patients with allergic rhinitis.
变应性鼻炎(allergic rhinitis,AR)是耳鼻喉科常见疾病,其发病机制尤其是基因调控仍不完全清楚。微小RNA(microRNA,miRNA)是人体免疫应答中重要的基因调控因子,对免疫细胞的分化、发展、调节起着重要作用。已有研究表明,γδT细胞不同亚群参与了Th2型免疫反应,并有着功能相反的调节作用。本研究拟以miRNA对γδT细胞的调控为切入点,通过AR中γδT细胞各亚群的表达及与Treg细胞之间的关系,探讨miRNA对γδT细胞的调控及其在AR发病中的作用和可能机制。以期明确AR中γδT细胞的miRNA表达谱及其对γδT/Treg细胞平衡的影响及可能机制,填补国内在miRNA与AR研究上的空白,对最终阐明AR的发病机理起重要推动作用,为AR治疗提供可能的新靶点。
变应性鼻炎(allergic rhinitis,AR)是耳鼻喉科常见疾病,其发病机制尤其是基因调控仍不完全清楚。微小RNA(microRNA,miRNA)是人体免疫应答中重要的基因调控因子,对免疫细胞的分化、发展、调节起着重要作用。γδT细胞不同亚群参与了Th2型免疫反应,并有着功能相反的调节作用。本研究以miRNA对γδT细胞的调控为切入点,通过检测AR中γδT细胞的表达及其各亚群的分布,明确了γδT细胞在AR患者外周血中表达增加且以VγⅣ亚群为主。同时筛选出6种差异表达的miRNA,hsa-miR-100-5p,hsa-miR-4772-3p,hsa-miR-504-5p,hsa-miR-6646-3p,hsa-miR-2115-3p,hsa-miR-4772-5p,提示miRNA对γδT细胞的调控在AR发病中起着重要的作用,填补了国内在miRNA、γδT细胞与AR研究上的空白,对最终阐明AR的发病机理起重要推动作用,为AR治疗提供可能的新靶点。
