The activation state, degree of infiltration, functional characteristics of T cells in the tumor microenvironment determine the results of anti-tumor immune response. T cell immune response in the tumor microenvironment is inhibited, the general role of immunotherapy is to restore the function of those T cells, but it will lead to tumor microenvironment immunosuppressive effect. In this study, the levels of tryptophan in the tumor microenvironment and the PD-1 / PD-L1 signaling pathway were analyzed which influencing the function of tumor-infiltrating T cells. The effects of IDO and PD-L1 expression in tumor microenvironment on T cell function were studied by in vitro and in vivo animal experiments. Explore the effect of combined use of IDO inhibitor and PD-1 monoclonal antibody in the treatment of breast cancer. Reveal the possible mechanism of this combination therapy in breast cancer and provide a more effective feasibility program for the tumor immunotherapy for breast cancer.
T 细胞在肿瘤微环境中的活化状态,浸润程度,功能特点往往决定了机体肿瘤免疫应答的结果。肿瘤微环境中的T细胞免疫应答处于被抑制状态,一般免疫疗法主要作用是恢复这部分T细胞的功能,但是这却会导致肿瘤微环境的免疫抑制作用随之加强。本研究结合两个影响肿瘤浸润T细胞的功能的因素:肿瘤微环境中的色氨酸水平以及PD-1/PD-L1信号途径。经体外和体内动物实验,研究IDO和PD-L1 在肿瘤微环境中的表达对T细胞功能影响,探讨联合使用IDO抑制剂和PD-1单克隆抗体治疗乳腺癌的效果,以期揭示这一联合治疗方案作用于乳腺癌的可能机制,为肿瘤免疫治疗乳腺癌提供更有效的可行性方案。
T 细胞在肿瘤微环境中的活化状态,浸润程度,功能特点往往决定了机体肿瘤免疫应答的结果。肿瘤微环境中的T细胞免疫应答处于被抑制状态,一般免疫疗法主要作用是恢复这部分T细胞的功能,但是这却会导致肿瘤微环境的免疫抑制作用随之加强。本研究结合两个影响肿瘤浸润T细胞的功能的因素:肿瘤微环境中的色氨酸水平以及PD-1/PD-L1信号途径。发现相对于PD-1治疗组组,联合治疗的效果并不明显,免疫细胞群体也无明显区别。我们发现相对于治疗无效组,治疗有效组小鼠T细胞pS6表达更强,表明免疫治疗能否有效可能取决于T细胞糖代谢是否被激活。紫杉醇相对于IDO与PD-1 mAb结合具有强大的抗肿瘤作用,可能低剂量紫杉醇能够刺激T细胞糖代谢,促进T细胞因子的分泌和T细胞抗肿瘤作用,并抑制了Treg的功能。
