Atherosclerosis(AS) is a common pathological basis of numerous cardiovascular and cerebrovascular disease, which caused a variety of cardiovascular and cerebrovascular diseases in today's society to become a major disease of death and disability. AS development requires two factors: atherosclerosis risk factors and the loss of arterialrepair function.Preliminary work indicates that herbal cake-separated moxibustion can fight AS risk factors by lipid-lowering, anti-inflammatory, anticoagulant effects. The new study has found in our team that the Herbal-cake-separated moxibustion could activate the expression of PPARγ protein in rabbit aortic endothelial cells to have the effect of delay the AS plaque formation. EPCs is a factor to repair vascellum ,and also a new target to treat AS. PPARγ can increase the number of EPCs, and improve the proliferation, adhesion, migration. Are herbal cake-separated moxibustion related to endothelia repair? The project intends to use herbal cake-separated moxibustion to AS rabbits as subjects, the use of cell culture, flow cytometry variety of biological techniques, ultra transmission electron microscopy, acoustic densitometry techniques to study herbal cake-separated moxibustion increased the number of blood EPCs and improve endothelial function in order to repair its anti-atherosclerosis formation mechanism, providing a new strategy for the study of the pathogenesis of AS and prevention ideas.
动脉粥样硬化(AS)是众多心脑血管疾病的共同病理基础,由此引发的各种心脑血管病成为当今社会主要的致死和致残疾病。动脉粥样硬化的发生发展需要两个因素同时存在:动脉粥样硬化的危险因子,动脉修复功能丧失。内皮祖细胞(EPCs)是促进血管修复的因子,是治疗动脉粥样硬化的新靶点。前期工作发现隔药饼灸可通过降脂、抗炎、抗凝等作用对抗动脉粥样硬化的危险因子,且能激活AS兔主动脉内皮细胞中PPARγ蛋白表达从而起到延迟AS斑块形成的作用。有研究表明PPARγ能够增加外周血EPCs的数量,并改善其增殖、黏附、迁移的作用。隔药饼灸是否与修复内皮有关?本项目拟采用隔药饼灸,以AS兔为受试对象,运用细胞培养、流式细胞术、超微透射电镜技术、声学密度定量技术等多种生物技术手段,研究隔药饼灸增加外周血EPCs数量并改善其修复内皮的功能从而抗动脉粥样硬化形成机制,为AS发病机制与防治思路的研究提供新的策略。
动脉粥样硬化(AS)是众多心脑血管疾病的共同病理基础,由此引发的各种心脑血管病成为当今社会主要的致死和致残疾病。前期工作发现隔药饼灸可通过降脂、抗炎、抗凝等作用对抗动脉粥样硬化的危险因子。本研究从隔药饼灸对AS兔内皮祖细胞的调节作用为切入点,观察隔药饼灸巨阙、天枢、丰隆、心俞、肝俞、脾俞穴对AS兔动脉内皮的修复作用。研究结果表明隔药饼灸通过提高AS兔血清SDF-1、VEGF含量,激活外周血生成EPCs,增强其黏附、迁移、增殖、血管生成能力而参与EPCs修复内皮的全过程,改善AS兔内皮环境,从而达到治疗AS的作用。且这种促修复作用优于直接灸。为AS发病机制与防治思路的研究提供新的策略。
