转基因沉默延长香石竹切花品种的自然保鲜期

The full length of ACC oxidase genomic DNA PCR product was cloned and 4 binary vectors were constructed, those are sense, antisense, direct repeat of sense and direct repeat of antisense. Totally 8 cultivars were used for genetic transformation mediated by Agrobacterium and 3 of which obtained transgenic plants. Comparison study showed that transgeic plant with sense foreign DNA can prolong vase life foe 2-3days while antisense construct can prolong vase life for 3-4 days。The results indicated that both the sense and antisense foreign gene can silence the endogenous gene at different level, different construct of T-DNA has different silence effect. Genomic DNA can be used to silense endogenous gene effectively. The cultivars with different performance of vase life are commercially valuable, the further study will be concentrated on comercialization of the cultivars.
