Oxygen convulsions, with the generalized convulsions as its most serious manifestations, may happen within short time since the commencement of exposure to O2 with 300 kPa partial pressures and above. With its severe injury to body, the underwater task and the clinical treatment with hyperbaric oxygen (HBO) are badly impacted. At present, the mechanisms are not fully understood and there are still no satisfied measures to prevent. Ferulic acid (FA) is main active constituent of Angelica sinensis, and in our previous studies we have verified it could markedly prolong oxygen convulsion latent period. In this program, we tried to elucidate if this effect of FA related with its effects on platelet derived growth factor-BB (PDGF-BB). The results showed that FA could notablely increase PDGF-BB content in brain vascular endothelial cells and neurons of cortex and hippocampus; PDGF-BB could significantly increased convulsion latency, and could protect neurons' form and functions from damaging by HBO. These results confirmed our hypothesis, FA could prevent oxygen convulsion through facilitating PDGF-BB production in brain vascular endothelial cells and neurons of cortex and hippocampus.
氧中毒是在江海深水处高压氧环境下作业人员的常见病,其首要表现为惊厥而危及人身安全。最近国外研究表明,脑血管中血小板源生长因子BB的增加可抑制惊厥发生。我们研究发现阿魏酸对氧中毒引起的惊厥有明显的抑制作用,本课题旨在研究高压氧下阿魏酸和血小板源生长因子BB抑制惊厥发生的相关性和定量关系,为阐明抗氧惊厥的机理和研究新药打基础。
