The spike-development transition in wheat from vegetative to reproductive growth is the critical development stage and determine the number of spikes and spikelets.The study of the spike-development transition can be further parsed the internal mechanism on spikes and spikelet formation.The spike-development gene TaFVE regulates the floral transition from vegetative to reproductive growth and plays the critical role in the biological process of the wheat tiller growth into the spikes. But the specific regulation mechanism is unclear. In our study, a spike-development inhibition wheat is as material used to identify the dowmstream target genes which are regulated by the TaFVE by the CHIP-Seq.Those target genes related to the spike development will be analyzed the transcriptional expression patterns by qRT-PCR. The protein factors which bind to the promoter and regulate the TaFVE are identified by EMAS and mass spectrum. Our results will provide the theory basis for studying spike development transition in the reproductive growth period.
小麦穗发育由营养生长到生殖生长转变是决定分蘖能否成穗和小穗数的关键阶段。开展穗发育转变的遗传机制研究能够为进一步的解析成穗数和小穗数形成的内在机制奠定基础。小麦穗发育基因TaFVE参与调控小麦穗发育转变进程,在小麦分蘖发育成穗中起关键作用,但具体的调控机理尚不清楚。本研究以成穗受抑制小麦为材料,利用染色体免疫共沉淀-高通量测序(CHIP-seq)技术和实时荧光定量PCR(qRT-PCR)技术分离和鉴定TaFVE基因调控的下游靶基因并分析其在营养生长到生殖生长进程的转录表达模式。利用EMSA和质谱技术鉴定与TaFVE基因的启动子区特异顺式元件结合的调控蛋白,分析转录水平调控该基因的分子机制。通过对TaFVE基因在穗发育转变中的调控作用研究,试图去揭示小麦穗发育的部分分子机制,为小麦穗发育遗传机理的研究提供参考。
