Pre-eclampsia is a specific pregnancy complications, and the major cause of the maternal and perinatal death.The existing studies suggest that the root cause of the disease is in the placenta, and a large number of studies have found that placental trophoblast excessive apoptosis and the pathogenesis of preeclampsia is closely related.Mitochondrial pathway is one of the classical pathway of regulating cell apoptosis.CYP11A gene is the key molecules located on the mitochondria.Our previous studies have found CYP11A gene over-expression in the preeclampsia plcenta,so,in the futere step,we will make a study of the relation between over-expression of this gene and nourishmentapoptosis.It has been found that abnomal androgen hormone secretion and insulin resistance is related to preeclampsia. The CYP11A gene encode protein P450Scc ,and the protein is a key enzyme in steroid hormone synthesis and metabolism.We will ,through specifically stimulate the expression of CYP11A gene in trophoblast cells, to research the generelation between CYP11A gene and estrogen, androgen and progesterone synthesis and activity changes, in order to reveal the relationship between preeclampsia and CYP11A.
子痫前期是妊娠期特有的并发症,是孕产妇及围生儿死亡的重要原因。现有研究认为该病的根本病因在胎盘,大量研究发现胎盘滋养细胞过度凋亡与子痫前期的发病密切相关。线粒体途径是调控细胞凋亡的经典途径之一,CYP11A基因是位于线粒体上的关键分子,我们前期研究已经发现CYP11A基因在子痫前期患者胎盘上表达增加,因此我们将研究该基因过表达与滋养细胞凋亡的关系,探讨其与子痫前期发病的关联;已有研究发现子痫前期的发生与雄激素激素异常分泌及胰岛素抵抗有关,CYP11A基因单一编码的蛋白P450Scc是甾体激素合成和代谢的关键酶,本项目将通过特异性促进CYP11A基因在滋养细胞中的表达,研究CYP11A基因表达改变前后雌激素,雄激素,孕激素合成及相关酶活性的改变,探讨它与甾体激素生成与转化的关系,进一步揭示与子痫前期发病的关系,为阐明子痫前期的发病机理、病理过程,开发以此为靶点的预测及治疗方法提供新的思路。
