本课题研究髓系细胞触发受体-1信号通路在脓毒症发病机制中的作用,及益气活血解毒法治疗脓毒症的理论基础。提出益气活血解毒的临床有效方剂化纤胶囊干预髓系细胞触发受体-1信号通路假设,成功构建盲肠结扎造成的脓毒症急性肺损伤大鼠模型,通过流式细胞仪、real time PCR、Western blot等技术手段从整体动物和分子水平观察化纤胶囊治疗脓毒症急性肺损伤后肺换气功能的改变、肺通透性变化、病理学指标以及髓系细胞触发受体-1信号通路相关细胞因子如TNF-α、IL-1 、IL-6、DAP-12等的变化,明确化纤胶囊疗效,从分子水平探索化纤胶囊治疗脓毒症的机制和深层次靶点,丰富益气活血解毒方药有效治疗脓毒症的科学内涵。
脓毒症因其死亡率高发病机制复杂成为重症医学领域的研究重点,对于脓毒症尚缺乏有效的治疗手段,故需要进一步研究治疗脓毒症的发病机制与治疗脓毒症的药物或措施,对降低脓毒症病死率具有重要的意义。在前期研究中,课题组已经完成了急毒、长毒、药效、药理和药代动力学等中药研发的基础实验研究,在临床中取得了良好的疗效,但未能从分子水平进行系统阐述其作用机制,通过本次实验研究:揭示化纤胶囊通过对脓毒症大鼠免疫功能、改善凝血功能、炎性因子释放的调控治疗脓毒症,并且有效降低了脓毒症大鼠的死亡率。通过研究髓系细胞触发受体-1信号通路在脓毒症发病机制中的作用,及益气活血解毒法治疗脓毒症的作用机制,揭示出化纤胶囊疗效,从分子水平探索化纤胶囊治疗脓毒症的机制和深层次靶点,丰富益气活血解毒方药有效治疗脓毒症的科学内涵。在重症医学领域开拓新的治疗思路,运用中西医结合的思想攻克了重症领域的难题。
