The green macroalgae Enteromorpha were focused on their negative effects because they caused the green tide before the Beijing Olympic Games. But Enteromorpha showed high nutritional value as early as in the "Compendium of Materia Medica" as a medicinal record. In Zhejiang province,Enteromorpha prolifera was regarded as popular edible seaweed and was exported to Japan. Many research on their growth and reproduction have been reported, but rarely on the phytohormone of the algae. This proposal intends to study the regulation effects of the phytohormone on the reproduction process of E.prolifera and their possible mechanism investigation. The main contents including: (1) To built up a set of of trace detection methods of the algal phytohormones based on the LC-MS and GC-MS techniques, to study the variation of the phytohormones on their different reproductive stages, and the relationships of the phytohormones and their reproduction stages, and the environmental factors of the light, temperature, salinity, nutrition et al. (2)The exogenous phytohormones will be used for regulation of the growth and reproductive processes,and different phase of Enteromorpha reproduction. (3)The auxin, gibberellin and other hormone-related genes of the E. prolifera will be cloned, and biochip techniques will be used to study the phytohormone gene expression and regulation mechanism during the different reproductive process, and their response to environmental variation. This study will be useful for the breeding and aquaculture of the E.prolifera.
大型海洋绿藻浒苔(Enteromorpha)在北京奥运前期引发绿潮,其生态危害受到关注。但浒苔具有较高的营养价值,早在《本草纲目》中就有作为药用的记载,在浙江沿海浒苔作为食用海藻广受欢迎,并远销日本等国家。目前对浒苔生长养殖进行了多年的工作,但对浒苔中植物激素相关的研究未见报道。.本项目拟研究植物激素对浒苔繁殖过程的作用和调控机理。主要内容有:(1)建立浒苔中LC-MS、GC-MS为主的植物激素微量检测方法;研究浒苔不同繁殖阶段的植物激素变化规律,植物激素与其生长繁殖过程及光照、温度、盐度变化等环境因子的关系。(2)利用外源植物激素调控浒苔生长和繁殖过程、繁殖过程不同生活状态等。(3)克隆出浒苔中生长素、赤霉素等激素作用和代谢相关基因,利用基因芯片技术研究浒苔不同繁殖过程中植物激素基因的表达规律和调控机制,及不同世代个体响应环境变化过程中的规律等。本项目将为浒苔的育苗和养殖提供科学指导。
浒苔生长繁殖及其植物激素相关研究具有较重要的生态和经济意义。本项目分别以浒苔成体、幼苗、孢子、不同繁殖来源浒苔为材料,较深入地研究了浒苔繁殖过程的生理生化变化规律,以及外源植物激素对浒苔生长繁殖的影响;建立和优化了浒苔中6种主要植物激素的LC-MS微量检测方法,研究了宁波浒苔的年度季节变化和植物激素变化规律;研究了几种外源植物激素对浒苔生长生理的影响,研究了营养繁殖和孢子繁殖来源浒苔对植物激素水杨酸的响应规律;研究了浒苔不同繁殖阶段转录水平的变化规律;利用转录组和蛋白质组结合的方法研究了水杨酸对浒苔响应高温胁迫过程中的缓解作用机理。共发表相关论文10篇,获得授权专利 1项。该研究对于深入理解浒苔快速繁殖及其机理,进行宁波特色浒苔人工养殖具有一定的参考价值。
