Grain filling is an important process in determining the ultimate yield of crops. And the dynamic balance of the biosynthesis, transport and assimilation of the photosynthate during grain filling is critically related to the grain yields. As we all know, wheat is a main crop in the north of China and the yield of wheat is of vital importance for the development of national economy. So we intend to investigate a new hybrid line of wheat, "Xiaoyan 81", using comparative transcriptomics and proteomics on its flag leaves, sheaths, stems and the developing grains during grain filling in the field. High throughput two dimensional IEF/SDS PAGE combined with MALDI-TOF-MS is the first choice for studying the comparative proteomics. Further investigation on the PMF unidentified differentially expressed proteins by tandem mass spectrum of MALDI-TOF-TOF or ESI-Q-TOF will provide elaborate information on the dynamic changes of proteins during grain filling. The tandem mass spectrum also facilitates the studying on post translational modifications which modulates the activity of most eukaryote proteins. Combined with bioinformatics, it is possible to build a metabolic network on the photosynthate transport system during grain filling of "Xiaoyan 81". Together with the photosynthetic funcion, our systematic and dynamic studies on the photosynthate transport system in wheat during grain filling will help us to understand the relationship between flag leaf, sheath stem and grian on different levels, so as to explore the molecular mechanism of grain filling.
小麦籽粒灌浆过程是决定其产量的关键时期,而灌浆期间光合产物合成、运输和籽粒形成之间的协调发展是小麦籽粒产量形成的基础。我们拟以大田生长的冬小麦小偃81作为研究对象,对其旗叶、叶鞘、茎秆和籽粒在灌浆过程中的动态变化进行系统的研究。以蛋白质组学和转录组学技术为主要手段,利用双向电泳分离技术结合MALDI-TOF质谱技术,高通量的分离和鉴定在灌浆过程中差异表达的蛋白质。对于一级质谱未能鉴定的蛋白,通过 MALDI-TOF-TOF和ESI-Q-TOF等串联质谱进行进一步的鉴定和精细分析,并通过二级质谱研究在灌浆过程中发生的蛋白质翻译后修饰作用。通过生物信息学方法,对鉴定到的蛋白质进行功能分析,建立灌浆过程小偃81光合同化产物运输的代谢图谱。最后结合光合功能分析在灌浆过程中旗叶、叶鞘、茎秆和籽粒形成在不同层面上的联系,探索籽粒形成的分子机制。
