There is an important effect of heat stress on poultry husbandry. It is significant for the researches on heat shock protein and there is a possible applications of the results to poultry husbandry due to the great role of heat shock protein in poultry heat stress. Single nucleotide polymorphisms of heat shock protein 70 (HSP 70) gene and their correlations to tissue expression in chickens were studied in the present researches. The completed sequence of HSP 70 was determined firstly and then global screening for single nucleotide polymorphisms of HSP 70 was carried out with single strand conformation polymorphism (SSCP) method, denatured high pressure liquid chromatography (DHPLC) and sequence analysis based on the aboved completed sequence. Homology analysis for the completed sequences from different individuals showed that HSP 70 sequence was very conservative, and SSCP analysis revealed that there was a substitution of from 34A to 34G in mRNA of this gene, and there was another substitution of from 276C to 276G in CDS of this gene. DHPLC and sequencing showed there were at least 8 SNPs for global gene of HSP 70. Gene expressions of HSP 70 in chicken muscles and livers were studied with ABI 7000 Real-Time Quantitative PCR Cycler. Gene expressions of HSP 70 in the above two tissues under heat stress and immulogical stress were studied with the same methods, and the correlations between HSP 70 genotypes and expressions were determined also. The results are significant for our understanding the relationships between HSP 70 gene structure and function. They may be used for the improvement of chickens' resistance to heat stress.
根据家鸡热激蛋HSP70基因的核苷酸序列设计引物,通过PCR扩增研究该基因的限制性酶切片段长度多态性(RFLP),分析比较热应激及免疫应激下不同RFLP型家鸡HSP70组织表达的差穑τ肏SP70作候选基因进行抗逆性定位、将其表达规律应用于家禽生产打下基础。该项目结果在育种方法的改进、提高家禽生产效率等方面均有重要意义。
