Insomnia has been a global problem and affects the quality of human life. Suan-Zao-Ren (SZR) decoction, a classical traditional Chinese formula, was originally described in Jin Kui Yao Lue, which has been used for treatment of insomnia, restlessness and anxiety. In the published, SZR decoction has been proved to nourish the blood and calm the nerves, eventually producing a tranquilizing sensation with high effective and low toxicity in the clinical practice in China. However, the efficacious material basis as well as the effective mechanism of SZR decoction has not been revealed. In this project, the experimental program was guided by an original Prescription of Traditional Chinese Medicine. The combination of serum pharmacochemistry, pharmacokinetics and metabonomics is adopted with HPLC-Q-TOF-MS、UPLC-MS/MS methods to establish the dynamic serum fingerprints and organism metabolic networks, which are used to compare the differences between constituents absorbed into blood from SZR decoction and its decomposed recipes. Based on “dynamic changes of constituents absorbed into blood - organism metabolic networks” strategy, Principal Component Analysis (PCA), Canonical Correlation Analysis (CCA) and Artificial Neural Network (ANN) are applied to reveal the relativity between the chemicals and metabolites, which will not only clarify the efficacious material basis and the effective mechanism, but also provide a scientific basis for the diagnosis and treatment of insomnia.
当今失眠症已经成为全球性问题,不断困扰人们的正常生活,给人们的健康带来极大危害。酸枣仁汤为《金匮要略》中经典方剂,具有养血安神、清热除烦之效,临床主治心神不宁、睡眠不安,但其药效物质基础和作用机制尚不明确。本课题拟在中医药理论指导下,采用血清药物化学、药物动力学、代谢组学等方法,结合HPLC-Q-TOF-MS、UPLC-MS/MS等技术,构建大鼠血、尿、脑整体机体代谢网络;通过建立动态血清指纹图谱,比较酸枣仁汤全方与拆方血清移行成分的差异,确定机体代谢网络中生物标志物群的变化,依据“动态血清移行成分-机体代谢网络”策略,运用主成份分析、典型相关分、人工神经网络等数理统计方法,揭示酸枣仁汤化学成分与内源性代谢物变化的相关性,阐明其药效物质基础和配伍机制,通过综合药效指标验证,初步探讨其作用机制,为酸枣仁汤临床应用提供科学依据。
本项目以《金匮要略》中经典名方酸枣仁汤为研究对象,对其有效部位进行了一系列研究。1.建立了超高效液相色谱-四级杆-飞行时间串联质谱方法,对酸枣仁汤体外化学成分进行定性研究,所应用的多种数据分析方法如质量亏损、子离子过滤等可大大提高分析过程准确度和效率。通过上述分析方法和手段,共鉴定101个化学成分,并将这些成分分别与五味药材进行成分归属。其中11种来自酸枣仁,9种来自茯苓,22种来自川芎,17种来自知母,47种来自甘草,有5种属于共有成分。这些成分按类别分类,可分为黄酮类、皂苷类、内酯类、三萜酸类、酚酸类、香豆素类以及其他类型化合物。同时解析了各类别化合物的典型质谱裂解规律,初步阐明了酸枣仁汤的化学物质基础。2.建立了超高效液相色谱-四级杆-飞行时间串联质谱技术结合多种数据分析方法对灌胃给予酸枣仁汤的大鼠血清样品进行定性分析。初步鉴定了35个原型成分,包括7个黄酮类、9个皂苷类、10个内酯类、5个三萜酸类、1个香豆素类、2个氧杂蒽酮类以及1个有机酸类成分。原型成分的代谢物经初步鉴定为33个。代谢反应类型主要为水解、还原等I相代谢反应,以及葡萄糖醛酸化和硫酸化等II相代谢反应。3.建立了超高效液相色谱-串联四极杆质谱技术分析方法,对大鼠灌胃给予酸枣仁汤后7种有效成分斯皮诺素、芒果苷、新芒果苷、阿魏酸、甘草苷、异甘草苷和芹糖甘草苷进行同时测定。血浆样品经乙酸乙酯-异丙醇 (1:1, v/v) 液-液萃取后,采用Venusil MP C18色谱柱进行分离,以0.1%甲酸水-甲醇为流动相进行梯度洗脱,流速为0.4 ml/min。所建立的方法用于对比灌胃给予复方酸枣仁颗粒和酸枣仁汤后大鼠血浆中7种有效成分的药物动力学行为研究。结果表明复方酸枣仁颗粒组和酸枣仁汤组7种有效成分的药时曲线趋势一致,复方酸枣仁颗粒组除芒果苷外,其他待测成分AUC和Cmax值与酸枣仁汤组相比均有显著性差异。4.建立了超高效液相色谱-四级杆-飞行时间串联质谱技术和多种数据统计分析的代谢组学方法,对酸枣仁汤干预PCPA致大鼠失眠模型的血清和脑匀浆代谢组学进行了研究。结果表明,在PCPA致大鼠失眠模型中有关潜在生物标记物均发生明变化。
