The tongue coating is the important basis of TCM syndrome differentiation and treatment .The research group's observation found that clinical tongue coating plays a vital role in the diagnosis and treatment of diabetes,and can correctly reflect the conditions of the patients phase characteristics.On the early and middle stage that no obvious complications in patients with T2 DM with thick greasy moss,accounted for 50%.The sscholars'study found that EGF、E-cad and TGF were involed in tegulating the formation of the coating .Thick and greasy moss in the expression of TGF-beta 3 is too lou,high expression of EGF and directly involved in its formation ,and increased E-cad mRNAexpression might be EGF/EGFR signal pathway is an important link in.The topic with thick greamy moss in patients with T2DM as the research object,from tonuge coating formation telated regulatory proteins of EGF、E-cad and TGF-beta 3 expression and its correlation with FPG,hick and greasy moss molecular mechanism of formation and the impact of hyperglycemia on its formation.
舌苔是中医辨证论治的重要依据。课题组临床观察发现,诊舌苔在糖尿病诊治中有着极其重要的作用,能够准确反应患者的病证阶段特点。早中期无明显并发症阶段,2型糖尿病患者以厚腻苔多见,占50%。学者研究发现,EGF、E-cad和TGF-β3均参与调节舌苔的形成。厚腻苔中TGF-β3过低表达,EGF高表达且直接参与其形成,而E-cad mRNA 的表达增高可能是 EGF / EGFR 信号通路中一个重要的环节。本课题以厚腻苔2型糖尿病患者为研究对象,从舌苔形成相关的调节蛋白EGF、E-cad和TGF-β3表达及其与FPG相关性,探讨厚腻苔形成的分子机制及高血糖对其形成的影响。
舌苔是中医四诊合参、辨证论治的重要依据。课题组临床观察发现,诊舌苔在糖尿病诊治中有着极其重要的作用,糖尿病患者早中期无明显并发症阶段,患者舌苔明显些以厚苔多见。学者研究发现,EGF、E-cad 和TGF-β 3 均参与调节舌苔的形成。厚腻苔中TGF-β 3 过低表达,EGF 高表达且直接参与其形成,而E-cad mRNA 的表达增高可能是 EGF / EGFR 信号通路中一个重要的环节。本课题以厚苔2 型糖尿病患者为研究对象,从舌苔形成相关的调节蛋白EGF、E-cad 和TGF-β 3 的表达水平以及其与FPG 的相关性,探讨厚腻苔形成的分子机制及高血糖对其形成的影响。课题组临床收取132例患者,分为糖尿病厚苔组、糖尿病薄苔组、健康人厚苔组、健康人薄苔组,留存其血清样本以及舌苔样本,行血清ELISA检验以及舌苔标本PCR检验,探寻EGF、E-cad、TGF-β 3 在血清以及舌苔中表达水平与血糖水平、舌苔厚度、血脂水平等指标的相关性。结果显示:健康薄苔个体血清中E-cad 、EGF 含量高表达;2型糖尿病患者血清E-cad表达水平较相应健康对照组增高,依然在糖尿病薄苔个体中高表达;2型糖尿病患者血清EGF表达水平与舌苔厚薄形成无关。血脂、BMI 与糖尿病相关,与舌苔厚薄无关。PCR检测内参GAPDH,以及EGF、E-cad、TGF-β 3 、TGF - α表达水平,结果显示:E-cad在糖尿病厚苔组与薄苔组之间具有统计学意义(P>0.05),在糖尿病薄苔中E-cad含量含量低,厚苔中含量高;PCR其余均无意义;同时发现基因于舌苔中含量较低,难以进行科学检测,结果可信度不强。课题组综合ELISA血清检测以及PCR舌苔检测发现,E-cad对于糖尿病以及舌苔厚度具有一定的差异性,可以进行深一步科学研究,以寻找中医药预防诊断治疗糖尿病的新靶点。
