Northern slat blot analysis was used in charactrization of fruit-specific expressed ACS gene in this study. Two fragments of the cDNA of ACS gene were ligated and cloned into pBluscript M13, designated pBS-ACC, to obtain the full length of cDNA encoding ACC synthase. The full length of ACS cDNA excised from pBS-ACC with restricted endonulease SalI and XbaI was inserted into plasmid pBBB digested with SalI and XbaI in the direction of 35S→3'ACS cDNA5'→terminator, from that the newly formed construct with antisense ACS cDNA was resulted . The construct was used in genetic transformation of banana. Apical meristems and corms from shoots of tissue cultured banana plantlets were used as material in genetic transformation via agrobacterium-mediated and microprojectile bombardment. Among the transformed buds, there were 64 regenerated plants resistant to PPT, among them 42 plantlet were analyzed by using PCR. The 9 plantlets were comformed to be transgenic plantlet and transferred to field to be ready for further examination for physiological features.
