"脾主时" 是五脏应时学说的一个组成部分。文献研究表明脾与时有"脾主四时之中"、"脾主四时之末"和"脾主长夏"三种不同相应的归类,均停留在理论思辨的层次;对于其现代内涵的揭示,缺乏系统的研究。脾为气血生化之源,中医防治疾病注重"未病先防"、"因时制宜",因此对于"脾主时"分化的研究尤为必要。申请者前期研究表明实脾法能增强AD大鼠海马中GFAP的表达;文献研究表明,钙蛋白酶(Calpain)与肌肉萎缩,及阿尔茨海默病等疾病的发生有关。因此,本研究采拟用免疫组化和生化酶学的方法,于"脾主四时之中"、"脾主四时之末"和"脾主长夏",分别探讨运用实脾法对AD大鼠GFAP和Calpain含量变化影响的规律,进而系统对中医理论中脾与时的不同相应归类的理论内涵做出科学的解释。从而补充"脾主时"现代研究的不足和空白,完善"五脏应时"的研究内容,对临床运用"脾主时"理论指导AD的防治提供现代科学的依据。
“脾主时”是 “五脏应时”理论的一个重要组成部分。脾与时有“脾主四时之中”、“脾主四时之末”和“脾主长夏”三种不同相应的归类,均停留在理论思辨的层次,对于其现代内涵的揭示,缺乏系统的研究。中医防治疾病注重“未病先防”、“因时制宜”。研究者前期研究表明实脾法能增强AD大鼠海马中GFAP的表达;文献研究表明,钙蛋白酶(Calpain)与阿尔茨海默病等疾病的发生有关。本研究采用免疫组化和生化酶学的方法,于“脾主四时之中”、“脾主四时之末”和“脾主长夏”,探讨运用实脾法对AD大鼠GFAP和Calpain含量变化的影响,研究结果表明在脾主之时,健脾益智法一方面可以有效的抑制AD大鼠体内TChE的活性,提高乙酰胆碱的水平,保护胆碱能神经元;另一方面通过升高大鼠海马区GFAP阳性表达,促进星形胶质细胞的激活,达到治疗AD的作用,从而验证“脾主长夏”和“脾主四时之末”理论的科学性。本研究通过这种“天人相应”观指导下的“以时测脏”的思路确切地证实脾经气血旺盛于长夏、四时之末,以及在脾脏功能活跃之时,“因时制宜”对AD进行治疗可取得显著的疗效。因此依脏腑相应之时用药很有必要进行深入挖掘研究,以完善中医学“时脏相应”的现代科学内涵及临床运用。
