The degeneration of cardiovascular system function in the process of human aging has become a key factor inducing major cardiovascular diseases such as heart failure. In the aging population, up to 40% mortality caused by cardiovascular diseases whose mortality rate was first among all kinds of diseases, seriously endangering human life and health. At present, the research on prevention and treatment of heart aging and heart diseases caused by aging is still insufficient, especially the regulation mechanism of heart aging has not been clarified. Therefore, it is of great scientific and clinical significance to elucidate the regulation mechanism of cardiac aging degenerative changes and reveal the occurrence and development of cardiac aging. In this project, we found that the expression of long-stranded non-coding RNA-DANCR was significantly up-regulated in the process of heart aging. DANCR-BNIP1 participated in the process of heart aging. This project will research on the regulation of DANCR and BNIP1 in cardiac aging by transgenic animals, explore the molecular mechanism of DANCR in regulating BNIP1 expression, reveal the role of DANCR and BNIP1 in the development of cardiac aging, and open up a new avenue for the prevention and treatment of cardiac aging and other degenerative diseases.
人体衰老过程中心血管系统机能发生退化,是诱发心力衰竭等重大心血管疾病的关键因素。在老龄化人群中,心血管疾病引起的死亡率已经高居各种疾病首位,严重危害人类生命健康。目前针对预防和治疗心脏衰老以及衰老引起的心脏疾病的研究还很不足,心脏衰老过程的调控机制尚未阐明。因此,阐释心脏衰老退行性变化调控机制,揭示心脏衰老发生发展过程,为衰老引起的相关退行性疾病的预防和治疗具有重要的科研和临床意义。我们研究发现长链非编码RNA-DANCR在心脏衰老过程中表达明显上调,参与调节心脏衰老过程。本项目将通过转基因动物深入研究DANCR和BNIP1在心脏衰老中的调控作用,探明DANCR调控BNIP1表达水平的分子机制,揭示DANCR与BNIP1组成的“信号轴”在心脏衰老发生发展过程中的作用,为心脏衰老等退行性疾病的预防和治疗开辟一个新的方向。
