Langerhans cells ( Langerhans cells, LC) are the main antigen presenting cell in the skin. LC remains immature in normal epidermis but becomes mature in some skin diseases. It has been found that haptoglobin (Hp) is resident in the cytoplasmic of LC and can inhibit the functional maturation of LC. In the present study, Hp expression in normal human skin and some skin diseases was detected by semi-quantity reverse transcription -polymerase chain reaction (RT-PCR), in situ hibridization, immunohistochemstry, western blotting, etc. It was found that there was correlation between Hp phenotypes and some skin diseases, and that keratinocyte (KC) was the Hp synthesizing cell in the epidermis and might be the only or main origin of Hp in LC. We also observed that Hp production in HaCaT cells was significantly increased by exposing to ultraviolet light. Our results not only elucidated that KC was an extrahepatic cell where Hp was synthesized and Hp phenotyes were correlated to some skin diseases, but also suggested that any of physical, chemical or biological factors that could lead to more Hp production by KC might have therapeutic value.
我们的研究已发现结合珠蛋白(HP)对朗格汉斯细胞(LC)的功能成熟和抗原呈递有抑制作用。我们拟进一步应用培养的小鼠的LC研究HP对LC的MHC-ll类抗原表达与代谢、协同刺激分子和细胞因子的表达等的影响,以阐明HP抑制抗原呈递细胞功能的机理。本研究不但可以加深我们对免疫皮肤病发病机理的理解,而且有助于我们开发新的治疗手段。....
