In view of the myocardial injury caused by ischemia, western medicine cardiac protection effect is limited. Prophase work confirmed that Danggui-Buxue Decoction had cardioprotective effect, but the molecular mechanism of its absorbed components is unclear. Acute myocardial infarction (AMI) is the important cause of myocardial injury, MAPK is a key pathways involved in cardiac protection. Literature reported that some active absorbed components could be confirmed after rats took Danggui-Buxue Decoction. It is reported that Danggui-Buxue Decoction and its active absorbed components has anti-atheroslerosis,lipid-lowering and endothelium-protective effects through MAPK pathway.Based on the above reported, The research will start to determine the absorbed bioactive compounds from Danggui-Buxue Decoction into bloof, then to apply the methods such as molecular biological skill and Liquid Chromatography with Mass Spectrometry to study the molecular mechanism of Danggui-Buxue Decoction and its bioactive materials in performing cardioprotection from the AMI-myocardial injury-MAPK-cardioprotection pathway based on the whole body, cell and molecular lever, and to quantitate the contribution of absorbed compounds spectrum into blood to the original formula, finally to open up a new way of research of cardioprotection by Danggui-Buxue Decoction and its absorbed bioactive compounds.
针对缺血导致的心肌损伤,西药心脏保护疗效有限。前期工作证实当归补血汤有心脏保护疗效,但其心脏保护作用的体内药效物质基础及分子机理不清楚。急性心肌梗死(AMI)是造成心肌损伤的重要原因,MAPK是参与心脏保护的一条关键通路。相关文献报道大鼠服用当归补血汤后血液中可检测到部分活性成分,文献发现当归补血汤及其吸收的部分单体成分可通过MAPK通路抗动脉硬化、降脂、保护内皮。基于以上报道本项目拟从当归补血汤进入血液吸收成分谱出发,应用分子生物学、液质联用等技术,基于整体、细胞和分子水平,从AMI-心肌损伤-MAPK-心脏保护途径研究当归补血汤及药效物质心脏保护作用的分子机理,定量血液吸收成分谱对当归补血汤疗效的贡献度,阐明当归补血汤及其药效物质心脏保护机制。
