Buyang-Huanwu Decoction created by famous doctor Wang Qing Ren of Qing dynasty is a famous prescription promoting blood circulation and tonifying qi. It is used to treat stroke and stroke sequela. Buyang-Huanwu Decoction has been testified clinically to be effective on cerebral ischemia, cerebral embolism,cerebral arteriosclerosis and coronary heart disease. What we have done is to study the effective ingredients of this compound and its mechanism of molecular pharmacology of nitric oxide's signal transmission of Buyang-Huanwu Decoction in cerebral ischemia injury and to probe into the concentration of Astragalas Glycoside A ,Ferulic acid ,and Ligustrazin in rats'serum containing Buyang -Huanwu Decoction .In order to study the mechanism of Buyang -Huanwu Decoction and the effective ingredients of this compound we have focused our research on the following respects :1. We have analyzed the contents of Astragalus Glycoside A ,Ferulic acid and Ligustrazin of the rats' serum containing Buyang-Huanwu Decoction by MS, Fluorometry and CE .2 After observing the effect of rats'serum containing Buyang-Huanwu Decoction on the growth of rats' neuron ,we came to find that some of the ingredients of this compound can promote the proliferation of rat carlical neurons, which indicated that there was some active substance functioning on the proliferation of neurons.3.We have probed into the effect of rats'serum containing Buyang-Huanwu Decoction on apoptosis of rats' neuron under the condition of hypoxia, and studied three important regulation moleculars in the route of Neuron apoptosis. As a result we have found that Buyang -Huanwu Decoction could significantly inhibit the neuron apoptosis induced by hypoxia. One of its inhibition mechanisms may related whit up-regulation of bc1-2 and lower-regulation P53 and P21. 4.We have also found the rats'serum containing Buyang-Huanwu Decoction had an inhibitive effect on nitric oxide and the oxygen free radicals of rats' neuron in case of hypoxia .5.By studying the effect of nitric oxide on neuron apoptosis in case of hypoxia ,we have found nitric oxide at high concentration was toxic on neurons.6.In the experiments we have found that Buyang -Huanwu Decoction could reduce the release of LDH, the level of nitric oxide and TBARS in culture medium indicating that it could prevent cardiomyocytes from apoptosis induced by hypoxia and reoxygenation .
观察补阳还五汤对实验性脑缺血性损伤的疗效与血清和脑脊液中黄芪甲甙和阿魏酸浓度的关系,在此基础上应用含药轿清和脑脊液培养神经细胞,观察其对缺氧神经细胞的凋亡程度、NO水平、nNOSmRNA和iNOSmRNA表达强度和NFKB表达水平的影响,及其对p53、p21waf1表达和OFOO-生成的作用,从而阐明该方疗效与其体内代谢动力学相关性以及疗效的深层NO机制。
